Just about to make some slants.

25ml of agar in a 50ml centrifuge tube.

How long at 15psi should I cook them for?

I did it for 90 minutes the other day and killed that agar and caramelized the malt extract.

Good job!  👍...you've come along way.

Is that from multispore?

The last Plate has a bunch of potentially good selections.

Hey I only select 1 from the last picture would you select more from that plate?

^ What he said. Good job on expanding into agar - seems you're on the right track to figuring things out.

Got some pans on deck.

Received a Zap culture that is taking off fairly fast on the first transfer.

And then I took the one little fruit from my current Tamp grow and put it on a plate. Plan on doing a transfer from this plate and then sending. See if I have any better luck from this rather than my LC.

Just the one and only early fruit that it threw. Still nothing on the respawn.
I'll let you know if anything happens on the respawn or from this plate once I send it.

Yes I would do several (some to the same plate, some to their own)

So almost every plate of agar I've had was bacterial when I thought it was clean. I think I may have finally got a good plate to go to grain with, how does she look?

This one I'm pretty sure is dirty. The red circle had a tiny white dot appear before the two sides merged and now that area is a bit raised. Where should I transfer from on the plate to best avoid contamination?

Here is the dot I mentioned before the merge. Surely that's not myc?

Looking at that 2nd image,  I'd go for 4 o'clock and 7 o'clock transfers.

First time doing agar work, using LME and golden teacher spores. 6 days after inoculating the agar this is the result. I know it's not much but the interesting part is the blue circle, the hair like strands of whatever is growing in the plate. I wish I had my microscope to see how it really looks.

At about what time should I start to transfer the good parts to a different plate?

After a few days growth those little islands of growth should have expanded to be big enough where you may see sectoring occurring. You want to grab a piece from the edge of the colonies when transferring to a new plate. 6 days germination time is standard.

Don't need to wrap HG plates. I've had a pour plate be just fine for 2 years with no infection. They're solid.

When they fail is when there's moisture pooling on the side, contams can swim up the water where they find...air swimming?...up the threads impossible.

I guess I didn't mean "unwrap", I meant that I had just taken them out of the jars that were in the PC.</font>

Oh, I feel ya.  I pressure cook by in the quart jars with the plastic white lids on em. Ill open them up in the hood when I'm ready.

Side note, I sterilize my HG's separated from the lids. Makes it pretty easy when I go to pour, as I don't have to worry about where to put the lids when they're cooling. Has worked out pretty well so far.

Okay, so should I be waiting until I observe "sectoring" or am I good to go to start transferring to a fresh plate? The room I keep the SAB in is a little cold, around 68-70 degrees fahrenheit, not sure if that makes a difference in how fast/slow the growth is.

Right on, getting an FFU is starting to become a necessity. I'm a lame-o and do my HGs as no-pour. But I now have LME and I'm going to start filtering my agar, so cleaner plates coming soon!

#1, horrible photo. Not much can be derived if we can't see what's going on. But in general, germination plates are usually iffy. Especially when new. You want to make transfers when you see something that looks healthy and homogeneous. You don't want to transfer sad wispy growth, you want something fairly dense and white. You may still grab mold thinking it's mushroom mycelium, but until you know what you are looking at, you won't know. Multiple transfers are your friend. From each promising plate, I would take 3-5 transfers to new plates. Keep looking at pics of good plates and compare to yours. You can normally only get clean cultures from lots of bad ones.
And temps are excellent.
