May I please borrow your eyes? These are “plates” I made 5 days ago. I think my agar recipe is a little too wet, or my dishes condensed. But otherwise… Thoughts? There are some pink spots in the first one, can I save a piece that isn’t in contact with it? Is the second contam? Some were wetter than others. And I know LM isn’t a cube, but does it look clean? Any advice on where to sample from? Thanks in advance for helping me learn to recognize what I’m looking at! 🙏

How are you making these? Whatever the container is seems non-rigid, which probably isn't doing you favors when you're moving them around.

Also, if you're taking the lid off and hovering the camera above to take pictures, unless you're doing that in front of a flow hood or FFU, you're also taking a very large risk of mold spores and other nasties falling from your phone/camera/hand onto the plate.

#1 has plenty of nice spots to choose from. Avoid the contamination (I see bacteria and what looks like another mycelial colony near the top edge).

#2 looks pretty awful. Might be some salvageable bits at the leading edge. Where it's growing over the edge of your agar puck on the bottom looks good if you can reach it without touching a bunch of other stuff.

#3 colonized for too long to give you a reading. Take a transfer from basically anywhere and see how that plate grows. Lion's mane looks a bit different than cubensis on agar, btw.

I know they aren’t pretty pours, but I had more agar than I had mason jars, so these were just poured into disposable sauce cups for a side quick experiment. The cups themselves were not sterile, so there’s that, but I didn’t want to waste the opportunity to learn, nor the extra agar. The agar was still a little warm so it warped the first two a bit. I don’t have a LFH, so I use a SAB inside a 4’x8’ greenhouse tent where I culture and cultivate (indoors.) I close the room up, wet down the inside of tent with soapy water, and use 70%, but that’s the best I can do right now. I was wearing gloves and trash bags, and ISO’d myself and my phone before entering the tent, but I realized it was a risk. I took it because I don’t know what I’m looking at and need the education from those who do. 😅

Ps. hoogshagenii for instance can get damn near black on agar.

My first time growing zapotecorum. damn thing kept going super yellow lol. i finally said fuck it and sent it. turned out great. now i see zaps go brown as well.

How do my plates of an isolated P. tampanensis strain on agar look??

Plate 1 has a noticably flatter, paler section extending outwards from the middle, and an outer ring of fluffier mycelium growing pretty uniformly around the perimeter (with the exception of the gap around 4 o'clock). What causes this growth pattern? Is it normal?

Plate 2 has a similar pattern to plate 1, with a patchy secondary ring of fluffier growth in the middle (particularly visible from 7 o'clock to 11 o'clock). Again, is this normal? Why am I not getting the pretty-looking rhizomorphic growth pattern I've seen from other isolated strains on agar?

Looks like tamps. Those rings are strange, no idea what the cause is. Maybe someone else has seen it before and can chime in.
Tamps don't really get rhizomorphic like cubes or some other species.

Only thing I could think is maybe moisture. Got fluffier as the environment dried out? The second one "spots" could have been form some last drops of condensation.


Total guess but all I could come up with. Inserting for sure.

I sometimes see the wispier central growth when I switch agar media recipes (e.g. MEYA to MEA - just omitting yeast - or to grain water). Curious whether it goes away on your next transfer to the same media.

It could also be a touch bacterial or have much genetic diversity - was this multispore or clone, and how many transfers out?

Clean tampanensis mycelium looks linear, sometimes a little aerial, never super ropey in my experience.

I'd transfer as far as possible from that certain contam at the bottom and from whatever that is on the left and middle.

So, top right, that rhizo to the right of the end of where you wrote the word "Zaps." I'd transfer tiny pieces there to several plates, maybe 3 or 4.

If you do that, grow it out and post your best of the 4 once it's grown out to about half this big and get feedback.

I think this will need 2 more transfers at least.



Thank you for your opinion, I super appreciate it.  I'm going to do a bunch of transfers tommorrow.  I'll update everyone on how it goes.

T1 of a clone I'm currently working with on 1/2 strength rye-water agar. This variety never fails to impress when it comes to rhizo growth.

Looking for some help interpreting some plates - I have a MS syringe from a well know vendor of a 'designer' combo of strains (B+ x Jamaican) that I cannot seem to get good looking rhizomorphic growth from.  This is a T4 transfer on PDYA:


T4 transfer because I've been trying a few different agar recipes.  It has since filled out the plate completely with 100% of what's in the picture. While that was growing I went back to a swab plate:




I have taken the leading edges of this to new plates - waiting to see what happens.  Please let me know your thoughts on these plates and how to proceed.
