Hi.

I cloned a GT and this is T1.  I took 4 different photos to try to show what I'm asking about. There is a thin "shiny" front running ahead of the growing myc.  I would appreciate help understanding what it is.

Here is the normal top view of the plate:



Here is a through view, if it helps:



Here I'm a little out of focus on purpose - if you look at the "glistening" part, you can see the shiny "thing" running ahead of the good stuff.  It is clear to my eye, but I have to hold the plate "against the light", if that makes sense, and then I see this part reflect light.  Hard to photograph.



This is as magnified as I can go:



Any ideas what this is?

Congratulations. You've found the leading tips of the threads. That's where it all begins.

If you're worried about it try transfering just a bit from that part and see what you get.

On the other hand it could also be bacteria growing slightly faster than the mycelium with the mycelium initially weakly rolling into it and then fluffing up as it grows over.

Looking for feedback & critiques. blue agar is BRF, red is LME, all 2%. should i be using karo? almost all T1 plates grew tomemtose in the center until about day 10. All PE, clone was taken from a plate pin. How soon should a transfer be taken from the plate pin?


the two T2 plates have some denser spots near the donor wedge. i've noticed this happens if it doesn't easily come off the scalpel, so i'm thinking that strands landed there. thoughts on the myc?

The exact same thing happened to my plates. The tissue clone was super rhizo and the T1 looked exactly like yours.

sweet comparison, guess the difference in growth has something to do with digestive enzymes.

had to stop cultivating for a while and was just studying & refreshing my memory to start over(agar).
Im no expert but that brf has vigorous & prominent - has both types of growth visible (rhizo+tememtose) to isolate.

just a guess,
brf/Grain water = more starch
LME = more maltose

Made some gnocchi (potato based) the other day, and the boil water looked thick and starchy, and since my grainwater agar has been underperforming, I figured this could be a good alternative.



I'll do 500ml, 8-10g agar and a tiny bit of sugar. I will also probably do 2 batches simultanerously at different concetrations.

Which dilutions should I test? Full gnocchi water, 1/2, 1/4?

This is an interesting read.

More or less says go full flavour with that potatoe water. I’d try it at full concentration and diluted 1:2. See which performs best. I suppose it depends what species you’re putting to said agar though as well.
