Can anybody tell me whats wrong with this culture? T1 Penis envy on light malt yeast agar. Ive already taken transfers from similar plates. I expected much stronger growth.

I have some Nec D. and Ps. Natalensis that just doesnt rhyzo. Is that normal? They just look mostly dense but circular:

Huh??

Don't over complicate things. Cubes don't care what grain they are on.

Nec D is a pan cyan. It won’t ever be rhyzomorphic.

Turns out plucking some of these aerial strands of mycelium to transfer was a good idea :takingnotes:

2.5 weeks later:

Gonna do a transfer and send this to grain

Are you not concerned about the scalloping?

:whathesaid:

I would not send that culture to grain with those scallops, do you have the original germ plate that came from?

The scalloping is a concern, yes.
But the other 85% of the plate looks better than the last few I’ve sent to grain and fruited without issues. Worst case I lose a quart of grains but still have the new plate
I was planning to take a transfer to see if the scalloping gets eliminated on the next plate and watch the plate and plate colonize simultaneously.

You guys think the scalloping on the edges make the plate bad enough to not send?

Rhizo isn’t inherently better than tomentose. Chase healthy growth, not just ropes. I mean chase the ropes if you want, just don’t prioritize it over healthy mycelium.

My best performing culture has transparently thin tomentose growth on the plate. Almost not even there it’s so wispy. Throws big fat weewees.

Anyone prefer to use tweezers or a loop to pinch/scoop the mycelium off their donor plates for transfers instead of cutting/punching wedges out?

I did this with two plates recently and noticed more uniform circular growth and may lean into it more if it’s preferred by some people.

You think a culture needs fruiting before it can be judged at all?

I have a project going at the moment where I mixed a bunch of different spores together, germinated them, selected the 15 fastest growing cultures on agar and the 15 most organised cultures on agar, now growing out all 30 cultures.

The theory I'm working with is that I want both the fasted colonisation and the highest yielding fruits. I figure the colonisation can be judged by how fast it colonises grain and highest yielding after harvest. The quickest fruiting perhaps too. But the selection on agar was very arbitrary, speed and organisation seems as good criteria as I could come up.

As far as we know those are good criteria. I mean on a plate what else do you judge? I’ve had great myc give me terrible tubs, so honestly how a culture fruits is really all that matters imo

Aside from needing another transfer (already done), does anyone see any problems with the myc on this plate? Penis envy t1 on 1.5% LMA.

My cultures keep getting an orange tinge about 2 weeks after spawning.
