How long do you wait to inoculate fresh plates? When it’s cool or do you give it a week to see if they are contaminated? My last go around I just left them in my SAB let them cool and gel then I inoculated them the same day. I have a plate from over a month ago and it’s still clean. Just looking to see what everyone else is doing. Currently making more agar rn to overcome the huge L that was my second go around.

They rarely contaminate so these days I allow them to cool and decondensate in laminar flow (a few hours to overnight) then use them

Considering my last plates all came out clean I’m pretty confident. I’ll only inoculate 2 to start and let those play out so it won’t be a huge loss.

Also how is everyone storing none inoculated plates? Are ziplocks fine just chilling out? Last time I put them in the fridge but I got a lot of condensation. So just wondering if I can leave at room temp in a ziplock.

Poured about 50 plates yesterday. Plans to use them all in the next few days.

Pushing out a bunch of Pans for some Fahtbags and to try out these 64oz containers.

I let them cool in the SAB overnight with the arm holes covered, use what I need, put the rest back in the sleeve they came in, and put them in the refrigerator.

I wouldn't store them in an unsealed bag in a fridge. Those are filthy.

Ziplocks in room temp work great

Silly question but bag them up inside the SAB right?

Lol yes. I can fit 4 per quart bag and that's perfect for me, just grab one and go.

If your confidence level is low you can always use fresh plates as germ plates, since you expect them to turn out dirty anyway.

For no-pours, I am stripping some of them as soon as they come out of the pressure cooker.

Check out these molds im growing; I am going to isolate the rhizo from the ape, but the country cock and equador I am uncertain. Should I just toss em and try again?

0x01) APE T1
0x02) APE T1

0x03) Equador Spores
0x04) Equador Spores

0x05) Nec D. Spores
0x06) Nec D. Spores

0x07) Country Cock Spores
0x08) Country Cock Spores

I ate my first gen ape, cleaned off the bacteria laden sugar agar; smelled like mushrooms after that. Before smelled dank and nasty

I didn't wipe my hands down with bleach before innoc. I also did not PC the agar long enough. Some of these have a hilarious amount of spores; each spore also carries its contams. so, ill probably use less next time.

How concerning is condensation in dishes? Poured kinda hot and one plate I inoculated is really wet inside I already wrapped it so don’t want to open it back up. I placed them upside down if that helps it but the water won’t evaporate because it’s wrapped.

Generally speaking I would say that some condensation is not a major issue, just flip the petridish over like you have done. If it pools then you need to be very careful moving the petridish.

It can make it difficult to observe the growth if it does pool as you have to lift it above your head to look into it without tilting it too much and risking the water getting onto the mycelium and the condensation can visually obstruct your vision also. Not sure how a very humid environment inside the petridish would affect the mycelium. Maybe the mycelium likes it?

I try to avoid condensation where possible so I use triple vented petridishes, lowered my agar % so I can pour the plates cooler, pour them as shallowly as I possibly can and leave them in a stack in laminar flow for a few hours after pouring them. If there is any condensation when I come to inoculate them then I allow them to sit open in the laminar flow for a few minutes to clear it. Usually while I am inoculating one plate I have the next one open to decondensate. If they are badly condensated before I start and that method would slow me down too much then I will open multiple in the laminar flow and decondensate a batch before I begin.

oh okay im currently just using a 110qt SAB so leaving them open isn't really an option I don't think...

the one that is really wet isn't pooling so that's good and I think the non-inoculated dishes are able to evaporate inside the ziplock so I think that's a plus... My last plates came out mint so this is a bit disappointing

It's not concerning so much as a pain in the ass. It happens. A lot. There are things you can do to try to avoid it:
-Pour as cold as possible
-(If you have a flowhood) let them dry in front of it, turning them part way
-(If you have a SAB) probably pour them and then carefully transfer them to another tote once cooled to fully dry before wrapping them, or just leave them in your SAB and wrap it...and wait...like a week or more.
-You can wrap them right away, kind of hot, and just wait (I know, the waiting part sucks)
-Upside-down works kind-of, and you can leave them upside-down and just pull the bottom off of the plate (which would be on top) when taking your transfers/wedges, leaving the wet mess on the bottom.
-Various methods of pre-pours with paper towels (I haven't tried this)

Note:
Unless the water has somewhere to go, it might move around, but isn't going anywhere. If it's clean, it won't be a problem anyway as it might catch and carry clean myc. If there's bacteria, it might give it a ride as well sadly.

Tl;dr -- don't worry, try again next time, and just use them as best you can upside down for now

Do you spray iso/soapy water inside the SAB? Perhaps if there is a very humid environment inside the SAB during inoculation it could make the condensation worse. Just a thought.

Do you know what you might have done differently this time compared to the previous time when you didn't get an issue with condensation? Was it the hot pouring?

okay cool.. only one is really wet so I think the ones in the ziplock will be fine come transfer time. thanks guys:heart:

and ya I spray the inside with soapy water, same as last time the only thing I could think of it was just a hot pour.

or it could be my heavy ass pours same recipe and I only got 15 plates compared to the 18 last time that could be it too more agar = more hot? idk

also if I bagged up the non-poured plates inside my SAB could I use them for later pours?

Yeah hotter and deeper pours will result in higher condensation so it sounds like that could be the cause. The more liquid you pour into a petridish the more water there is to be released during cooling.

Consider shallower pours. I make 330ml of solution and pour 20 plates with it with 5-10% of the solution left over at the end that I discard. So approx 15ml per 90mm petridish. It only half covers the petridish when pouring and I have to move the petridish around manually to get the liquid to cover it.

Pour when the agar solution is about 50 degrees Celsius (or as low as you can get away with).

Re: AGAR ENVY! (Anything and All things agar!) [Re: covertjoy]
    #28456374 -

ya I let gravity and the agar do the filling which 100% leads to heavy pours.. I guess I could pour a few and then rotate them around to help even it out. Working within the SAB is what makes me move quickly and not want to stop pouring once my media bottle is open. once I can prove to myself I can get fruits (and have space to have a clean work area) I'll be more willing to cash out on an LFH

I know not necessary but im clumsy and I think I would benefit greatly.

I pour that shit thick, let it steam while I smoke a j, then bag em.  No struggles with condensation if you pour at the proper temp
