I'm trying out my new transfer tool. 16g needle, bevel ground off, 3 cc syringe with paper clip on the plunger to push out the sample. Diameter is 1.8 mm, the narrow side of the transfer.  This is APEU, T1. The spore donor is the result of many agar and plain transfers.

I've yet to try glass plates, but I feel like the extra weight would fuck me up if I went 100x20.

Wish they had 60x20

They have
60x15mm
100x10, 15, or 20mm
150x20mm

Would the 100x15 or 20mm really be too much? It does seem like the smaller ones are more popular (back ordered).

I'm sure it's something you'd get used to, I just can barely fit both lid and plate in one hand

I've been hoarding alot of cups, its gotten into the hundreds and I need to throw some away. I've got a ton of cryovials full of sterile distilled water for the first time. I've got my MVP cultures set aside. I have all my clones separated and original germ/early generation cup for each when applicable. I'm in the process of organizing them still and its hard to let go, but its time. I just need someone to tell me if I'm forgetting anything important before I throw them out.

If I have say a t2 or t3 from a pin clone or germ plate, isnt that just a clone of a clone and unnecessary really for long-term storage? I'm trying to preserve heterozygosity and save myself time later so I've been saving every transfer and I've been keeping track of filial generations from spore, clone, transfer #, and can trace each line back to its original predecessor. What are the most important things to keep honestly?

its been a few years since ive used a real petri. I think ill pour some 100s this weekend before I decide.

how long you looking to store things?

any transfer from a clone should be the same as the donor plate.
i would keep the top and bottom of every generation given its clean. if that makes sense?

a spore plate transfer could be wildly full of traits tho. what are you really trying to preserve?

well a few of my cultures have extremely dense pinsets and very high "cupin" rate. Many of the pins aborted, but it was like a carpet of pins, no visible sub. I want to preserve that trait. I also have some clones and 2nd filial generations that are displaying prolific cluster and high rates of growth. Some are dense canopies, some are small, some are huge. The clusters are 40-60 pin clusters.

Im not sure i follow. keep clones that retain the traits you want and toss the ones that don't.
I hate to give you such generic cloning advice since you seem to be on the right track, but there is a limit to how many pins a sub can support, and they've gotta have room to grow.

are you trying to just store cultures long term or are you trying to change the traits of a lineage? realistically what do you need to keep, yaknow? its hard to articulate the point im trying to make. Im sure i lack the vocab

Has anyone ever seen this? A bright pinkish dot has appeared in my dish. Was going to be taking transfers from this guy but now I think I’ll use one of my other plates.

Just take your transfer on the other side of the dish, away from that pink spot.
I think that myc looks funky tho, but it might just be the picture quality.

That’s always a pain. If anything, try not to overthink it. It *could* be a thousand things.

Verifying major ingredients have been used with success by others helps. Especially stuff like your grain and your coir. If you’ve done that, I’d take a deep breath and go back to spore if possible (I or many others could help if you need new study material). Keep running what you have with patience but as much as possible simplify.

Good luck! I’ll stay tuned to your journey!

You can't see viability like that. Only way is to try and fruit it.
Could always just take it to a small cake or bottle tek and attempt to tip fruit to save some time
