Agar question

What is the consistency of colonized agar? Like when you cut a triangular piece out of it and stab it with a knife to remove it from the tray, is it similar to stabbing a piece of apple? Or is it sorta runny and you have to scoop it out?

Agar should NOT be runny, nor should it break when removing it from the petri dish. Soft and pliable best describes good agar consistency.

It really depends on the % of agar you used and the temperature you are working in. I use agar for plant cultures (8g/L agar), and I find that if I leave the plates out they are softer, and fall apart more easily.
For mycelium, the recepies I see call for 20g/L agar. Your plates for mycelium should be pretty solid though. Try to keep them cool and out of any places too warm as the agar will melt.

15 g/L works pretty well. It depends somewhat on the quality and source of your agar. I like mine a little on the soft side and it saves a bit on the agar.

You can check out my media cookbook listed on this page...

Just remember to change the recipe for how you like it. 15g/L is good for pretty much all of them. Harder or softer work better for different situations.
