Agar - Inject or just drop solution
    #7049366 -

When working with Agar are you supposed to inject it?
Does this work better. Or just drop solution on it?
When I drop it smears all over the plate when flipping and
my plates got all funky.

After I pour my plates I let them sit till they congeal + a day.

In a glove box i either:

Drop one drop of spore solution

-OR-

Dab my spore print with a loop and make a Z shaped swipe across the agar.

When done I wrap in Parafilm and put them on my shelf at room temp.

After colonized I take the best sector (about the size of a freckle) and put on another dish.

Hope this helps

I'd recommend letting the spores sit on the plate upright for a few days before flipping. This will give them time to stick to the agar and not flow around with the syringe water. The only reason to flip the plate is to reduce condensation on the lid.

This helps alot and thank you. Is there a max time you leave your plates in incubation? Basically trying to figure out if there is no
healthy growth when should I toss.

Thanks workman. I think I have been pouring my plates too hot
because they have been condenstaing like mad. Even though I put a
hot cup uf water on top.

I leave my plates for a couple of months, but the longest delay in germination I have seen is 21 days from dry spores. Typically I see germination in 4-10 days depending on the species and age of the spores.

Condensation is a problem that you just have to live with. You can reduce it by pouring cooler agar, stacking plates as you pour and incubating at a constant temperature. Temperature fluctuations increases condensation. But as long as its not so bad that it causes flowing water its just an annoyance.

Don't pour your plates until you can hold the bottle of agar with latex gloved hands without burning yourself. If the jar/bottle is too hot to hold, it's too hot to pour. If you wait until the agar has cooled, and then stack your dishes vertically, there will be ZERO condensation on all but the top dish. The condensation on the top dish can be eliminated by placing an old or empty dish on top. It's the temperature differential that causes condensation. stacking dishes vertically stops that problem.
