without seeing the spawn it’s just guesswork, but the only realistic cause of contams when spawning to coir is gonna be unclean spawn. coir has been known to contam on its own but it’s really unlikely imo. those tubs look bacterial & it’s really easy to miss bacterial symptoms on grain spawn. snag some pics of your jars after a 100% shake next time just in case this happens again

i was more suggesting that just to be more confident that the myc was clean but yeah if you’re pheno hunting or trying to get prints i’d think the genetic diversity would be a plus. i mean as long as nothing too funky is going on in your dishes no reason to not send a t1

No it's not a scam, sectoring is somewhat irrelevant. Uniform growth is an indication of health. Each "sector" is made up of many tens of thousands of individual hyphae, maybe even hundreds of thousands. Separating them with a scalpel is impossible.  Much of the hyphae have fused to exchange nucleic information which blends genetic information. The idea that a cultivator is losing genetic diversity by making multiple transfers to achieve pure culture and uniformity is not very accurate.

By inoculating haphazardly with erratic growth you're exponentially increasing the risk of contamination, not preserving genetic diversity.

I was thinking I would make transfers to keep working on my agar skills and put the remainder to grain instead of binning if they look contamination free?

I started this journey to grow a supply to microdose for mental health purposes (with the added benefit of learning new skills along the way!)

You think these look totally fine to put to grain? Have some jars ready to go.

Gave a friend a few of my colonizing PF jars, they ended up soaking said jars for 4 days after colonization (they forgot about them), leading to foam, gas (plastic container was sealed ended up bursting open) and a sour smell from the containers in which those cakes were being soaked.
Are those cakes done for, or can they still produce fruit if they are cased? Other than the smell, the cakes look fine.

If your intention is to isolate genetic material for whatever reason, you're better off not allowing ms to anastosmose in a bundle. True isolation can only be achieved with single spore isolation or dedikaryotization. By the time your ms solution germinates they're in such close proximity to each other that anastomosis happens almost immediately.

The monokaryotic phase is the shortest lived for that reason, the preservation of individual strains is impossible without intervention.

im not trying to be a hater or anything, i would love to see someone come up with a super affordable flowhood design. the problem is, there have been a million β€œteks” on cheap flowhoods & they all end up the same. not saying it’s impossible for one to work, but no one thus far has proven a design that is reliable. you absolutely need more data & solid evidence that what you’re doing is as reliable as an SAB or traditional flowhood. just trying to save you from the impending shitstorm clusterfuck that you’ll have posting your tek with that amount of data. that’s great if it’s worked reliably for you so far, but we are gonna need repeatable & reliable evidence to believe your claims because i promise you people have tried this before & failed

you can get lucky a few times, or have something kind of work, but for something to be taken seriously it has to prove just as reliable as the baseline methods we tend to recommend (ie. an SAB). the shroomery is a goofy website at times, but at its core is an amateur science community & therefore it is expected that there is a reasonable burden of proof for anyone making claims that shake the status quo on here

multiple harvests will take time, so does good data. you need to really prove that this works reliably, every time, to good success or else your post will be picked apart to oblivion. amass a lot of solid data, run some really rigorous testing (doing multiple blank transfers, prove grain jars won’t contaminate when manipulated in flow, post pics of healthy subs that don’t contaminate during any point of their life etc.) & then post results. one agar plate alone is not even close to enough to prove anything to anyone on here



Thanks Hazy. You are much more familiar with the forum and norms than a newbie like me. Perhaps I was too enthusiastic with my early success. As you suggested I will step back and compile more data.
Getting picked apart doesn't faze me, I grew up  with an older brother :grin:

That’s what I kinda figured. It’s what I tell people. My limited understanding of β€œisolating” genes is that it isn’t something you’re going to do with just a scalpel and a Petri dish, so I honestly haven’t bothered learning. It’s a skill way outside the scope of my goals.

Follow up question: does wild sectoring always/usually indicate a problem? I’ve sent myc that wouldn’t stop scalloping after 5-6 transfers early on that performed normally. Is erratic growth sometimes just that, or is it caused enough by problems that it’s just better to avoid it?

I’m not being very clear. Is there a way to tell the difference between genetic diversity and problematic growth at a glance, all things equal?

Scalloped plates have always resulted in problems for me. The scalloping persists no matter how many transfers. When sent to grains the growth is always slow and unhealthy. Rarely do they ever fruit, when they do my tubs are filled with metabolites and excessive moisture. The fruits are very few and sickly, any fruits are a rarity even when top fruited.



These are a great example.

Even/uniform growth may take 3 or 4 transfers to achieve but is worth it; but plates like the ones above seldom produce anything workable.

Can you alter your substrate to fight a contaminated grain jar?

I read that increasing the acidity level of your substrate, makes it less hospitable for contaminants.

Assuming you have a slight amount of contamination in a fully colonized grain jar, would using a more acidic substrate help fight off the contamination?

As CocaineBuffet stated, the short answer is no. You could increase the acidity of your substrate using some sort of weak acid (or make it more basic with a weak base) to make it less hospitable to contaminents, however, it would also do so for the mycelium.

Stipe, do we know why this happens? I got some plates too that didn't all come out right. Arranged from "best" to worst, I mean the last one just completely gave up on life.



The "A" plates (first two) came from this one:

bottom right

I couldn't find the "B" plate, but here's C to F


The "E" is the most fucked up and like you said, it did produce the same fucked up child too.

So what do now? I'm thinking keep transferring from the first 4 and put the last 3 in the "let's see if it plins" pile. :shrug:
