Clone culture. Is that contam at 3 oclock? Hoping its just some residual myc that fell off there somehow.

If you didn't touch anything there or swipe the scalpel blade through there then I'd avoid that side in your next transfer cause it's a better chance it's a satellite contamination growth.

Should I be looking to transfer it though? I thought I should be wanting to keep and preserve all the genetic material I can?

The point of transferring is to get to an axenic culture. If that's a satellite contamination you don't have an axenic culture. If that were mine I'd watch that 9-10 o'clock region as it grows out a bunch more then take my next transfer from there.

I will surely do a transfer or 2. Just curious about the process of cloning and keeping all the genetics. Thought to successfully clone you got to get it clean first time in order to not transfer out any potentially beneficial genetics. Is this not true? Obviously if it is contam its screwed anyway.

You dont transfer out or in genetics doing agar work. You can't isolate genetics on agar. It's millions of mycelium filaments all woven and twisted all over each other. The difference of what can happen on a dish is how some of them form and grow.

I thought thats how isolating a monoculture/isolate on agar works? By narrowing down genetics.

Nope even a multi-spore culture can look perfectly symmetrical with no sectoring. And no amount of transfering will get you to any kind of an isolate or monoculture.

Even clone tissue can still contain lots of different genetics. Certainly a ton less than starting from spores. But still lots like 1000's instead of millions.

This is old misinformation we see and correct again and again. Probably from Paul Stamets. Or Reddit. Where did you hear it?

Agar transfers are used to clean up cultures (get a mycelium culture with no bacteria or mold contamination), and select strong looking culture, not to isolate to a single thread. And to clone (from a small section from a shroom). Clone cultures are always more narrow / predictable and often stronger than multispore (MS, that is, from spores).

What rumfor69 said is true.

Mycelium can't isolated to a single rhizo thread anyway. Someone here tried that to about the 80th generation, it kept making new multiple threads.

If you want to get good genetics with agar, grow some shrooms, select the best one (tallest, strongest, best looking, first growing, thickest, etc), and clone that, then clean it up, and grow it.

Read my sig line post "new, start here", there are some links in that post to some great teks on working with agar, cloning, and making transfers.

Stros cleaning and agar mentions mono cultures and transferring too much being counterproductive I think. Dont really remember exactly. Just was under that impression from what ive read. I only get grow info off this site. I realize theres a lot of outdated info.

Stro's Cleaning and Isolating on Agar is very solid for learning to do this, and it's in my sig line post I recommended to you.

But it's almost 10 years old,.

Except a few things (most of which I link in that post) I usually recommend finding teks under 5 years old on here, and when I do advanced search I limit to last five years.

Older teks often being replaced by newer info is why it says at the top of old threads and posts here "Some of these posts are very old and might contain outdated information. You may wish to search for newer posts instead."

An old tek that's mostly solid can have info that's no longer considered solid.

It is commonly repeated that transferring too many times is counterproductive, due to senescence (dividing cells degrading with age and many generations, part of the reason all living things age and die), but I think it's overstated, and was more overstated in this hobby in years past than recently.

Several TCs have said that's overstated, and they've transferred many times and still had strong shrooms grow robustly.

My argument for senescence being way overstated in this hobby is that there are a number of clone-based varieties (like Enigma) being widely shared that are incredibly strong are probably 200th generation clones or older by the time people get them now. Those clones are still going strong, grow well, and are some of the strongest cubes in the Psilo Cup for the past few years.

I just re-read the Stro's Cleaning and Isolating on Agar tek (always fine to have a refresher course), and I see nothing it it about too many generations degrading quality. It does say though "it is important that you don't get too wrapped up or concerned about 'sectors'"

Pic tax and example for the "there are no monocultures" idea, this is a plate where I transferred a piece of agar with a single thread of rhizo from a clone culture. It grew into a bunch of threads.

I've heard (from Sidnee Obersingleton on FB - he's a mycologist) that a clone only consists of a single genetic individual - that it's not possible for different cultures to cooperate in creating a fruiting body. At least not within the species we're mostly interested in here.

Hello guys , its my first time trying stuff with agar , and for sure that GT its weird af with that  yellow slime up in there , I have isolated a small part of mycelium to a new agar plate , but besides the GT does the rest look ok ?

Terrence is from spores and the rest are cloned from fruits.

Thanks in advance.

True or false: mushrooms (or other organisms) that develop without light will produce less pigment.

I thought this was true but I don't know the name for it. If it is true, how much lighter will mushrooms from the dark be?

Edit: thought this was quick questions quick answers. Here's a plate pin as tax for the agar thread
