First agar work from 26th of October. This is from spore syringes, am I growing mold or should I transfer some? 10/25 plates are looking like this and the others show no sign of growth whatsoever. Thanks!

What causes a recipe to fall out of suspension in the PC cycle?  Or, is that normal?

I used 10g/10g/1g MEANY with 500ml of water.

It's typically the yeast causing all the sediment since it's not water soluble. I preboil my agar/ME/yeast and zap it with an immersion blender before bottling and cooking it. That helps with the flocculation a little bit but you will still get some sedimentation towards the end of your bottle

Bigfoot isn't wrong about NY, I don't use it because it's not water soluble.

I completely disagree with the notion that agar doesn't have to be pre-boiled. My comment from several pages back spurred a reply that pre-boiling is 100% unnecessary, that it's like bringing sand to a beach. At the time, I thought it was good advice and said I would try it. (Don't know if it matters, but I'm doing pre-pour in jars; GWA only.)

I tried it twice, and it failed twice. I have since edited my replies, as well as posted a new reply, but it was swiftly ignored by all. I had to make a new batch last night, and I did NOT skip the pre-boiling this time. I didn't change any amounts of ingredients used, and the agar did not stratify.

I guess skipping the pre-boil works for some, but not everyone. To each their own, but I know what works for me.

What was your water level like with respect to the agar in the bottle when you did it without pre-boiling? I think you need the water to sit at around the same level as the agar, I.E. partially submerge the bottle

But I don't know. I'm a pre-pour guy

My agar bottles are completely out of the water in the PC. I started pre-boiling before I started using yeast because it gave me more clear (clearer?) plates and it's just a habit I haven't dropped since starting to use yeast. Other than some yeast flocculation I've never had any issues with stratification to my knowledge :shrug:

I'm not sure how to answer that, so I will just describe the steps I took.

I combined 100 grams of grain water with 200 grams of plain water plus 6 grams agar agar powder in a small cooking pot. I stirred until all the agar powder was completely wetted. At this point, the powder was not dissolved, just wet and clump-free. It stays settled on the bottom unless stirred, and quickly settles back out as soon as you stop stirring.

When I attempted to skip the pre-boil, I stirred constantly while sucking up the solution with a syringe, (no needle), and added the same volume to each jar. (20 mLs) Visually, each jar had roughly the same amount of agar powder before loading in the PCer.

The agar turned out looking like this: (The stack of lids is just to hold the jar at the correct height for the picture. Do note the dark layer at the top...)


For last night's batch, nothing was changed except for the fact that I heated the same cooking pot while stirring constantly. As soon as it was at a full boil, I removed from heat and continued to stir for another minute. (I noticed the agar appeared to be fully dissolved right before it reached a full boil, so I probably could have removed it from heat about a minute sooner. I think the key is dissolving the agar with enough heat, but a FULL boil is probably a little overkill.)

I immediately used the same syringe to add the same amount to each jar, although I did drop the amount to 15 mLs each jar. Loaded into the PCer, same heating times, everything else was done the same. The agar did not stratify this time at all. It resulted in a very clear and proper looking agar.

I believe I know why my agar did not set correctly when skipping the pre-boil, but I have already covered my theory in past comments.

Water level was at the level of the agar in the media bottle per Bod’s tek.

I’m going to make another batch and pre-boil this time.  If it stays in suspension, I have an answer that will work for me.

I don't do no pours but I imagine that you would have to preboil for that or you wont get even agar distribution.  You also don't have to boil, only raise the temp above ~70C to dissolve the agar.

I pour because it’s easy and I have 400 dishes to use up.

My first pour I had no issues although I had flakes of NY in my plates.  No worries, the myc didnt care.

This last pour, I strained water over the NY as a test.

The only other difference was pre-boil vs no pre-boil ahead of PC’ing.

I am going to test straining again with pre-boil to see if that makes a difference in the suspension of the solution.

I was considering buying some MP tape, but looked in my cabinet first. I had medipore and durapore tape, so I looked into how they compared. I couldn't find any specifics on the pore size of any of them, but I did learn my on-hand options aren't worth anything in mycology.

When I looked up the specifics on 3M MP tape, I was surprised to read this:

What size microbe is blocked by Micropore tape?

The term "microporous" may be used in several ways:

Edit: I also see that the hole may not have been completely covered by the tape. That one edge looks like it is not sealed due to the gap in the lid.

I dont use NY and dont preboil. 500ml and 10g/10g MEA, a good shake, and perfectly clear every time.
