I prefer the cheap shit ones honestly. They are a little thicker and the closer tolerances on the Pyrex causes an occasional vacuum lock.

The gap on glass plates are a little tricky to wrap at first but you get the hang of it. Instead of wrapping the side middle you wrap the middle of the pararfilm into the bottom corner at a 45 degree angle. I need to add that detail to the tek but it's hard to photo that without help.

Ahhhh reminds me of my old Tandy 1000 and my 486...Yes I am old enough to remember coding in the ancient MS-DOS days with QuickBasic, Turbo Pascal, C/C++ IDE, Assembly Language, and of course debug.com. ASCII art was your only GUI restricted to 1, 4, or if you were lucky 16 colors. Thought I would give props to those who remember computers&programming before drivers and plug-n-pray when you had to do all the low and high level stuff yourself. Sorry to be off topic but it brought back memories when programming was truly fun...and now back to your regularly scheduled thread. :grin:

I'm still on that no pour glass petri, I do them in large pp containers with massive polyfil stuffed lid, 10 petris at a time and leave them to cool inside the container.
When I'm ready to work I open the large pp container in front of the flow.

I've been prepping dishes and leaving them for 3 weeks in the pp container and all traces of condensation dries off.

Great tek sandman, I followed this to the letter for my first time pouring agar in petri’s (been doing no pour since I started). I think they turned out okay! I think I poured them a little thin but I guess you get used to how much to pour. Kinda hard in an SAB to see. This was the end result: PDYA (first time with this) and a pinch of activated charcoal

Just trying to get clear on how to store poured plates that aren't going to be used for a while.

I've been wrapping poured plates which is a nuisance as I have to unwrap then wrap again after inoculation especially as one of the reasons I got into glass plates was to avoid plastic waste.

What other options are there for storing poured plates pre-inoculation?

In zip lock bags but I still think wrapping them is safer either way.
Or you choose pp containers that have a good lid seal.

I just wrap em until i need to use them. Then i just grab what i need. Tried a bunch of other methods over the years and have settled one just wrapping them.

Do you use the same piece of plastic to wrap it back up? Not sure if it would have a higher rate of contam at that rate or if you can just wipe it down with alcohol prior to starting.

I did a batch of no-pour glass petris the other day and wrapped them after letting them cool and dry in front of the flow hood.

I then stored them in a tote box but on checking them today I noticed that many of the wraps have slipped off, underneath the plate.

Has anyone else experienced this? Note that the tote box hadn't been moved or bumped over the last couple of days. The plates were seemingly dry when I wrapped them.

I used grafting tape to wrap. Maybe I stretched it too tight? Maybe I wrapped it too low on the plate to cover the gap?

i just use seran wrap for mine & it sticks super well. if they are wet it’ll slip off but idk i’ve never used grafting tape. seran wrap works great tho

Yeah, I'll give saran a try. It's really weird how the wrap just slipped off without any handling of the plates. Maybe temperature shifts and some moisture and me wrapping them too tight and low?

I've rewrapped them and will leave for a few days to see it they contaminate.

yeah sometimes the wrap placement makes a difference with those factors but it is weird they slipped off. not familiar with grafting tape really but you shouldn’t have a problem w seran wrap

Yeah the point is, the dishes must be dry. Otherwise the clingewrap won't hold propperly.

You can buypass that prob with just using Parafilm :laugh: It's just much more expensive compared to clinge. I like to use clingewrap when wrapping up dishes that I store in the fridge for later usage. The dishes I cultivate in are wrapped in Parafilm (as long as I have, if it's out I also use clinge)

Cling wrap is for the kitchen. Parafilm is for the lab. I've never had that problem with parafilm.

I've left a batch of foil-wrapped plates in the oven to cool down.

How long can they be safely left there? They are double-wrapped in the foil and had two hours at 430F.

My oven isn't that clean so am concerned about whether leaving them there until I get home is a vector for contamination.
