What do you think about this oyster clones i did 5 days ago. I dont know if its oyster myc or just mold

How much time should i wait to see white mycelium in germ plates? Have some that can see a weak ramifications growing on the germ point bot not sure yet if its cube mycelium either.

So I'm new to the whole agar game... Some of these are made with MEA that I got pre-mixed from Amazon (Seaweed Solution Laboratories) and some is using PDA that I've mixed up myself (30g Dried/powder Potato Starch, 10g Agar, 2tsp of corn syrup, 1L distilled water). I'm thinking I might need a better recipe as the Potato ones aren't showing much growth (I'll hunt down some dried dextrose from the local homebrew shop next time).

I'm thinking 1L distilled water, 30g potato starch, 10g Agar, 20g dextrose. Does that seem like a reasonable recipe or should I be looking at something else? I need to stay away from Malt as I have a Celiac here and that's just asking for a different kind of contamination issue.

Second last pic is where I'm currently storing my plates. The last pic is my new laminar flow "hood". I built it myself and so far it seems to be working but I've read people saying they need to be tuned?

For the MEA ones (made on the 10th) are they healthy? I'm not great at identifying contamination yet, re: newb.

I made up some grain water plates 8 or 9 days ago and put a drop from a vendor syringe onto each.  The tops of the plates are a huge mess of bacteria but 4 or 5 days after I made the plates I noticed some spots that were either in or underneath the agar.  I checked in on them again today and the spots are definitely growing, and I'm wondering what I've got here.

Those are the clearest pics I could get of 2 of my plates but they all have similar spots on them.  Is that spot in the middle of the first pic some myc?  Did it just grow down through the agar to get away from whatever shit is growing on the top of the plate?  The kind of weird thing is that the spots are all over the plate and not just under the drop that I initially put on there.

If it is myc there's no reason not to try and transfer it, right?

I started my LAGM grow with a bacterial syringe with similar growth and took some transfers:

The myc eventually grew away from the bacteria on the T1 plates:

adhoc said:
Yeah I don't have high hopes but I'm going to let them sit around and see what happens.  I wonder about myc not traveling through agar though - isn't that how sandwiching and hot pouring work?

Did you end up taking any transfers? and here's a quote from RogerRabbit on hot pouring and myc growing through agar:

RogerRabbit said:
The hot agar partly 'pasteurizes' the bacteria which slows it down, while leaving the mushroom mycelium unharmed so it can grow up through the new layer.

The one on the right looks good. The left looks good from what i can tell, but is harder to evaluate due to the granular surface behind it. Oyster myc IME colonizes flat, fast and fine. not so ropey and rhizo.

Good luck!

Does anybody know how this happens? The bacteria is all on top of the plate.

I kinda want to grab some wedges but I’m not sure the middle is even clean :uhoh:

condensation may have collected near the edge and grabbed some nasties there and rolled it around a bit. Thats what it looks like to me.

I got a plate that's got since trich that I got from a friend. I want the culture, I the it in my fridge to slow down the green. I assume if I use my hood it's going to blow that shit everywhere, I figure I'm gonna have to bring my SAB out of the shed so there's no current. Anybody ever try to save a culture from the green monster using a hood with good results???

If myc doesn't start doin weird shit on it, I may have found my new favorite food coloring line. This black held up really well in the PC. I haven't found an interest in charcoal because I like to see through my plates. This is dark enough that it looks black but not so dark that you can't see through it. Kind of reminds me of grape Kool-Aid. This stuff can be picked up at hobby lobby and isn't very expensive. There's quite a few colors to choose from and their red's don't contain erythrosine.

The myc ate some of the food coloring on my black plates. Turned a forest green color.

Edit: different brand though. Plates look nice πŸ‘Œ

Myc always turns my plates a tan/pink color. These plates have a blue tint if you look close enough. I won't be surprised if they turn green.

Hey Mario, why'd you get the big hood and how much was frieght shipping? Just wondering. Awesome alternative to charcoal though btw.

I hate busting out my sab for a dirty plate, so last time I just I turned my hood off. Then I just worked in open air right in front of my hood like the room was a big sab. I figure just from moving the plate alone, everythings gonna be dusted in contam spores anyway, so why bother with the sab. I had about a 70% success rate, suprisingly. Probably would've been higher if I had parafilmed the plates and been less reckless but I expected mad contams so I didn't bother.

edit: to clarify, the hood was turned off after a session, so it was already running for 2+ hours. so the idea was the air immediately in front would be cleaner than the rest of the air for the next few minutes. but I've had good results just working with my hood on full blast too, even though it feels wrong. you can always beat the contam spores that landed there by just xfer'ing again before they germinate

I had planned on getting the 2x4 but was also considering the 3x4 because I had read some comments by a few people saying that the 2x4 was nice but they wished it was a bit bigger. I knew the FFU's were coming back in stock soon so I checked the website a few times a day. I checked again one night when I went to bed and it was all still out of stock. Woke up the next morning and checked again before I went to work and there was one 3x4 and one 4x4. After shipping the 3x4 wasn't too much more expensive than the 2x4 so I jumped on it and here we are. The FFU itself was $800.00 and freight was $280.00. It would have been a couple hundred dollars more but I had it shipped to the nearest SAIA terminal and picked it up myself. It's a bit excessive for what I'm doing but the space doesn't get wasted. Sometimes I like to stand in front of while I work instead of sit.

Thanks man, no way do I have the space right now unless I sat on the floor criss cross applesauce but I'd like to get one someday for supplemented sawdust bags for oysters. I thought shilling would be a lot more, good idea about the terminal.

Yeah, I really think I could've got 90% success easily. I only did 10 plates, so only 3 went bad with absolutely zero fucks given. no regurts

Put a β€œdrop” from 4/6 purchased LC syringes to agar 1/22/21. This is 2 of them 6 days on. The photos of Lions Mane and Shiitake won’t upload for some reason. The Shiitake drop is showing no sign as of yet. The Blue Oyster and Red Reishi both got more than a drop as the needles of both had resistance as I attempted to aspirate said drops.

Blue Oyster

Red Reishi

Not sure what to be expecting of these visually. Seem a bit disorganized. I have 1400ml of LC broth for each of them but wanted to confirm a  clean culture first.

I've got a few plates of APE going and I'd like some input. So far this variety is completely different than anything I'e worked with and has me scratching my head.

MSS -> agar 1/1/21 It's all chewed up because I cut my transfers out of this. The 'clean' colony on the bottom was a second spot that germinated on the plate.

T-1 plates As you can see it's that thin 'chalky' lookin' myc.

T-2 plates Still 'chalky' and slow as all hell.

So, I'm wondering if once this grows out a bit more would it be good for grains? If not, I'm debating on if I should take a transfer from a BRF puck and see if it either speeds up or looks more normal.

This is what my PE looks like after transferring from a BRF puck, and recovering from T-2s being taken from it.

Clear plate porn
