That's not what I'm saying either.
I'm having a hard time getting thoughts to text today.

all im saying is the side that isn't "leading" tends to recover better because its more established.

^this is all I'm getting at.
Take from the leading edge, make sure your culture is clean my guys, but once that is taken care of, if you're expanding a culture out onto enough agar dishes to drop onto grain you might as well take from where its well established.

These were leading edge transfers, you can tell where the leading edge was.
If I had fathomed this growth pattern had to do with how established my myc was I would have taken different sectors.

Interesting I’ve never paid attention but I will now.

I was always told you take the leading edge because that’s the newest most active myc growing and looking for food.

I've heard the same thing, along with the "its the cleanest myc" or "farthest from contam"... im not knocking any of those things... they are all visible in practice. If you did a josex style poke and put it on a plate you'll get close to perfectly round growth... when I was a noob taking 1cm squares id see it all the time tho... the leading edge still does fine and in most cases will catch up to the rest of the myc... it just seems that the more established myc on the piece your transferring will jump off better than the newer growth.

Hopefully I haven’t shared these two yet

Probably post a few more, catching up on some agar work next few days

Id wager that the side with the strongest growth is what would've been towards the inside of your punch... we talk of taking the leading edge, but the back half of that leading edge tends to have stronger myc than the actual leading edge.

Next time you take transfers, note which direction you place the "true" leading edge of your xfer. Ill bet ya a pack of spore prints the opposite side of the leading edge will grow stronger.

Thanks!  I'll start tracking the lead edge.

Does streaking plates with a swab usually end up in separated colonies like that? I've only ever used a print to loop, but I've heard it can be difficult to get spores off of a swab when streaking which would make those colonies make sense. Just curious :awehigh:

Edit: To clarify I'm asking if spore swab to agar will leave satellite colonies more often than loop to agar, because I've only ever gotten solid "z" germination from print>loop>agar. It would make sense that the swab would deposit less spores, and If you did a Z-streak it almost looks like the spores germinated on the angles of the "Z". Is this generally how swabs germ on agar? I ask because I have APE swabs I'm reluctant and scared to fuck up.

I believe that is the case... I also tend to streak a ridiculous amount of plates in the hopes that the later plates I swab will have less and less colonies, thus making them easier to differentiate.

When I streak print>loop>agar I get isolated colonies that look similar to these plates. I usually end up getting all the visible spore clumps off of my loop within the first ∼30mm of streak, so the rest of the plate has good isolated colonies. With swabs, you just kinda have to trust that the spores took the agar because you probably won't see any spores on the plates, just a z-pattern that looks like you swiped a blank q tip on them.

Btw, I'm not sure if these would be considered "satellite colonies"
When transforming bacteria cells, satellite colonies are colonies that haven't up-taken a plasmid forming around a colony that has. Essentially hijacking the antibiotic resistance ability of the larger colony and living in its protected environment, trying to sneak into your project and ruin it  :begger:

Some pics of plates made from 6-7 year old prints. The blue agar is the newest batch.

In regards to transfers growing out differently on different sides.... I have been growing my plates with the intent of colonizing the actual agar throughout. I pour very deep agar, wait til it fully colonizes the agar and make extra thick LI. If I grab some agar from these thick plates to make a transfer, I notice that the top surface continues to expand as rhizomorphic mycelia, but the inner agar is tomentose. The transfer people are taking near the edge might have one side with leading edge surface rhizomorphic growth but no agar colonized yet... and the more central side of the transfer having some of the agar colonized.

I was referring to those APE swabs so I'm glad you answered. I'll probably do 20 plates just to make sure :whip2: I haven't fucked with swabs so I'm going to try all the streaking, snipping, burying tricks I've seen :crazy2:

@ Doobsker, I was half a bottle of Jameson in so satellite slipped out heh, but I didn't know that differentiation so the Jame was lookin out. Thank you for that info :happyheart:

Can someone help me understand how, if at all, the spores from a huge canopied fruit differ from the spores of a single small one of the same variety.(different cultures)

Also interested. Heres a link about crossing strains, but I'm also curious as to when a line of genetics becomes "stabilized" and replicated by it's spores.

Until recently I thought that going back to spores was a complete genetic reset, but recently I'm seeing things that insist otherwise.

This is my first attempt with swabs.  Ban Phang Ka.  I jammed a tip of one swab into the agar on one plate and steaked the other plate with another swab.

Both plates took 5 days to display mycelium.  The tip showed and continues to display more robust growth than the streaked plate.
