When you have a jar full colonized, just extract a grain with some flamed tweezers and put it to agar, then make the bulk.

Glass Petri are getting easier. . .

If you can PC pre-pours (aka no-pours) for 15min, then you can sterilize empty petri in the same time? Bod's PC tek doesn't necessarily agree due to air pockets.

I generally PC stacks for 25 min and have no problems. . .

If you're lucky and own a Flowhood, you can leave your pour or pre-pours open to dry. . .

We use our bathroom (not toilet) for sterile sab work. Just never leave anything unwrapped in there and close any windows, ventilation before work starts and that's good enough, so it seems. Then again Im a lazy fuck and use premade industry agar plates. They come at about eur 13,- per 20 poured plates. If any get contamed b4 use they send replacements too :smile:

Sticking to the side of the jar?

I use this method. (sorry not the best diagram)

I just tilt it end over end then lay flat then, twist the jar left and right till grains start to swallow the wedge as its rides the grain then sit it right up. Haven't had agar stick to the lid or side since discovering this method of motion.

It's not the stickiness of the wedge but the motion of the ocean.  :derfase:

-edit- you can add more agar to your mix to make it more stiff as well, also you don't need to shake the shit out of it after a wedge drop, unless you use the SAGAR method. I have seen an increase in speed just dropping a wedge in and getting it were I want vs shaking it. It actually seems to take off faster because it didnt have to recover after being all beat up after the wedge drop.

That's what I do. I just slowly tilt the jar and spin it until it gets where I want, then I shake it good, then I get it back where I want. Done.

But today they all mostly decided to stick to the frickin jar

For me it's not so much those things, as just easy of use. I currently have 4 different PP5s I'm working with to determine the best for my uses, because they aren't available in too many varieties in the sizes we like (believe, I've spent fucking days, looking at all of them).

I like the idea of pour, PC and forget about'em. I've just been trying to find lids that work.

One of the 4 I have are the ZL extra-small, which I'm pretty stoked to use, because I love screw top and know they won't melt. They cost me 80 cents a piece and will last long enough to pay me back.

And then there are my deli PP5s and they cost next to nothing per container. Two are sturdy, but the lids are fucky and the last one has weaker walls, but great lids.

They all vary in size, so the one set are 93mm at the base, 85mm, 60mm and 57mm from there. I plan to use the smallest and weakest, to do my innocs and then move up in size per transfer to the largest. I have them now, so why wouldn't I use them?

Plus side, my wife likes having unlimited access to tiny containers for her lunches at work.

It's not that I can't pour plates, it's just that something about the lack of effort required after modifying the containers, to have a huge wack of ready to use agar is sexy to me :naughty:

For me the process of pouring in the SAB is laborious and often painful because of my back, but with no-pour I can line up a hundred plates on my counter and just bang them out. I also like the idea that once they're PC'd they're good to go. Right now I can only get 20 plates at a time quickly, whereas a 500 plate order wouldn't arrive till August and also looks shady as hell going to a residential address. ZL twist-n-loc ain't so shady :smirk:

I'm doing this to look at spores guys, who needs that many plates?:shrug::mushroom2:

I wish this could be the case in my house I have a single window unit to cool the house and its on the fritz plus nowhere near the lab. I have to have a fan going to maintain 83F in there that's not to mention the atleast 3F hotter it gets in tubs.
We slumming it on this side of town bro. I don't even know what central air is.

Lol I feel ya gizmo i got up at 5am and the growroom is 86 with the air on low. Between the constant air and the humidity dropping to 4% I think a lot of my tubs got fried. A couple shoeboxes look like I dropped a brand new brick of coir in without hydrating. New brand though too. :shrug2:

What's going on with these plates? I'm still quite new to agar, probably no more than 80 plates done in total.



Calling the top row plates 1, 2, and 3 left to right, bottom row 4, 5, and 6 left to right. These are all transfers from a fruit I cloned. Agar recipe is 20 g/L agar, 16 g/L light malt extract.

Plates 1, 2, 5, and 6 showed promising sectors of thick rhizomorphic growth early on (I guess plate 2 still does) but that seems to have given way to fainter, wispier stuff, with a pronounced halo between the first stage of growth and the later one.

Is this just a crappy looking (but faster growing) phenotype being expressed/taking over? Or something else I should be worried about?

If it wasn't for the contam I'd be fairly happy with plate 4: despite not being thick and ropey it's nice and regular.
