Ohh i wanna be on the agar thread too! I've never posted in this one.

PE6 clone looking a lil funny. Not thrilled with the wispy-ness of it all.

Some TOC a new one to me.

And some uber slow, cottony, and apparently deadly APE

I made that transfer the same time as the other two above. I have two more plates of it that are even slower. Those two are LMEA, this one here is grain soak.

I wanna say the wispy bits of the pe6 is mold.
The TOC has a mold halo, visible on the right. I'm betting it's moving pretty slow.
Ape looking good.

Yeah the PE6 is 1st transfer from clone. Theres no mold on the TOC though, its all pretty up close with no halo. I think that's a little condensation shadow on the lighted pic. The TOC plates are cruising  :stoned:

Hello there, my first agar test, both are some LC left overs, can i save something? i made some grain to agar and they looks good but will not open them until the transfers.

For my education, please let me know how that TOC does on grain/bulk.

I see the halo he’s talking about but so far I think it’s connected to the way the myc grows and how hard the plates are.

But I’m also just now finding more hidden contams in my jars that I didn’t know what I was looking at before. Now I have to back track to my plates and try to see if I can find something hiding in there or if it’s from bad sterile technique.

has anyone ever tried using polypropylene petri dishes in the pressure cooker? i have a couple coming in so i could do some no pours. my concern is, will it boil over? Also, how long should I pressure cook it? 45 minutes like pasty plates?

The polypropylene (pp5) petri I looked into were all autoclavable.
You only need to PC for 15mins (I do 20) for liquids including agar.
If they are like glass, use a bit more water in your PC to slow the cooling process and don't vent.
I'm also pretty sure you have to pour them at some point. . . So I call them pre-pours :shrug:

I’ve just wrapped mine in stacks of 5 with foil, pc and then pour... to me “no pours” are the same amount of work (if not more) but much more condensation to deal with. Never got the appeal

So far I'm with you D3monic. Last try at PCing plates came out sooo watery.
I had the trivet jacked up on a steamer, only ran 20 min. and sooo sloppy.
Now I'm just letting them sit in the SAB to see if they can dry out.

No pours can work, but the condensation is a nightmare to figure out.

These days I'm all about the no pour. I have surprising luck nopouring with 1/2 QT ziplock containers (great for pizza balls). Suck to view through, but I like to streak 3 lanes or drop 3 diff transfers. Then grow out this MS agar to clone the best fruits.

Here's some clean T2's on the huge plates. I like these different clusters and don't bother isolating.

Wow I feel dumb. I'm so used to pasty plates and no pour that I didn't even think about PCing the petris without the agar and then pouring in a SAB.

I stopped having condensation problems (like the third time I ever ran any) with pre-pours when I started taking them out as soon as the pressure dropped and setting them up on the refrigerator for a couple hours.  I use both jars and Ziploc containers and don't have all these condensation problems...  Only time I had any issue with it was when I was playing with moisture content of brf/rye/cornmeal pucks when I was out of agar.  Not really even the same issue, but I digress.

So are you putting petri plates in a jar?
And in a ZipLock/Rubbermaid containers in the PC?

I found a 32oz jar with 130mm opening, but they only say PP, not PP5.

Ouuwee, you can bake glass plates too. Foil, 400, 2 hrs, I think.

Do you tend to isolate further? Currently I just drop a whole agar like that on grain and grow a box and isolate from there.

Historically no, once it’s clean I take a little transfer and grain the rest and just kinda keep doing that. So it is getting isolated more each time.

This one just has an unusual amount of sectors for a clone t3 and they’re all nice and clear.

I’m curious though if there are actually way more or if it just grew in a way easy to see them. It’s a newer variety not far removed from the wild print so maybe there’s way actually way more diversity since it hasn’t been bred constantly for a decade.

It was also a pain in the ass to get a solid culture. Working from spores I ended up dropping wedges that I wasn’t even sure were myc, then it took three transfers from a clone to get solid growth all around which is crazy ime. With clones I’ve had beautiful plates right off the tissue half the time.

This was the spore plate.

I learned today that you don't need substrate to get shrooms! :smile: just posting cuz its the biggest agar shroom ive had so far! Took transfers from this germination plate a while ago, but I just kept it for shits and giggles!
