But I get the same contam rate (10% too much I know )
whether I use PP from a sleeve or my freshly baked Corning Borosilicates.

Thanks Poisoned, I think we had this conversation befoe...mebbe someone else.
But I could try again, I'm sure no pour would reduce my :horrified: contam rate!
Of course this time I could try PCing with the foiled stacks of plates above the water! :stars:
Also for extra Noobie credit, last time I wrapped a bit with cling wrap, so they stay together. But the cling wrap melted to the plates. I think I've finally scrubbed the last off of my beautiful plates. :heart:

YES! Those are by far my favorite plates. So strong and sturdy. I love those. Theyre more expensive than the budget plates you find for $50 or $60 a case, I think theyre like $120 a case, but theyre worth it imo. And they come in 10 pack sleeves instead of 20 or 25, so you dont gotta pour a whole 25 if you just need a few at that time.

I'm about to pour some plates with some old grain soak water.

It's been in a gallon jug with a filter disc RTV'd to the top and it smells like it's probably started to ferment a bit.

Does anyone know if it's still ok to PC and pour up some plates?

Second question, I've had B+ and GT plates going, growing pretty fast on MEA.

I put them to rye grain and the temp has been between 68F-78F in the room. I guess I was expecting maybe 1 - 2 weeks for full colonization.

I'm at week 2 with maybe 30%, no contamination but seems slow.

would I be better off selecting for cultures that grow quickly on grain soak agar to have a better chance of fast colonizing times?

I gave up with WBS grain water agar, contam rate was out 100% even with 60min PC; went back to MEA and no regrets, crystal clear with no contams!

AA+ grows out slow and thick. Older dish seems to have started blueing. Is this good, bad, or neutral in the way of myc on agar?

All of the above... slow, thick, the bluing, the lack of characteristics resembling cubensis mycelium, the lack of uniformity in structure... I'm not sure that plate is cubensis even... its not healthy imo

Second look and consideration of the blue/green... you MIGHT have penicillium on your hands... are you fruiting these cultures yet?

Seems any alcohol would evaporate during sterilization: give it a shot!

Finally just dumped a couple hundred old plates out so that I can stick em in the recycling.

I'm trying out my pp reusable petris along with my glass ones. Plan to just PC them and pour em after.

I'm not a fan of waiting for no pours to cool down inside the PC. Call me impatient (cause I am)


I hope they work out well. But I also just ordered a case of the 100mm Celltreat grip ring plates cause DAMN those suckers are convenient.

What are your objectives for each transfer?

Transfer 1 - transfer away from the unclean inoculate
Transfer 2 - transfer away from possible hidden contam

After you are sure it's clean from the first 2 transfers.....

How much do you let the mycelium grow out for each transfer afterwards, when do you start looking for sectoring/isolating, and when do you let it fill the whole plate to inoculate grain? Also, what do u do with clean plates from earlier transfers? Throw em?

Seems like most people leave unused clean plates alone, and if they pin you can clone it. Never done it before, but I have some on a shelf now to see if they pin!
