Is my liquid culture ok?

Hello, 12 days ago I inoculated a liquid culture that I bought online with 2ml of spores from a spore syringe. For the first 9 days there was no growth so I have placed the vial in an incubator at around 27°C. The day after the white cloud shown in the pictures has appeared. It looks like the spores finally have germinated but there are no mycelium strings forming, just white flakes. Is this normal? (in most of the pics I have seen online the mycelium looks like a cotton ball, not like little particles)

Shouldn't really innoc lc with spores as they're probably a bit dirty. Test it on agar and you'll know

You.wont know until you use the LC if it is clean or not. But the good news is you'll know pretty much immediately. If there is even a little.clean growth on whatever.you.inoculate then you're good. Looks pretty normal to me.

Visually, it looks great. But unfortunately, there is no way to tell if the LC is clean by looking at it. You have no idea if the syringe was actually sterile. Also,
by adding heat, you have upped your chance for any contamination which may be present to flourish.

All in all, it may work out for you :shrug: But you haven't set yourself up for success so far. Test what you have on agar. If you aren't in a position to do that,
you shouldn't be doing LC's in the first place...because you will ultimately end up wasting a lot of your time and materials for nothing.

Thank you for the answers, the problem is I can only get my hands on spores syringes and where I live agar is quite expensive, 30€/100g. So I tried grains with no success and now I have attempted LC. I guess I'll inoculate something and hope for the best.
P.s. should I wait or is the LC ready?

It's ready just swirl hard to break up the mycelium and then draw it up for use.

I would let it grow out a little more.



Hoping for the best rarely works in this hobby. Instead, I would prep some small containers of grain - an inch or two deep in a pint jar for instance -
and test with that. If that looks good, you can do your big jars.

It looks barely grown, most use their lc's when it's pretty much full without stirring.

Keep looking, there's gotta be some cheap agar source you can get. You're not the only european grower here, and they seem to find agar alright.

Spores are typically dirty to some degree, without agar, there's not much point in trying lc's and your grains will fail frequently(successes most likely bacterial). You'd have more success making brf cakes instead until you get into agar.

/e if it's the same syringe/batch that failed in grains, I would not be hopeful about that lc. LC is a more hospitable environment for contams than grains.

Just Google "telephone agar agar" should be around 10 euros for a quarter pound which will last for a few hundred plates or so.

Should also mention that 100g of agar, assuming you are making something like MEA (50% agar, 50% malt extract) is enough to make a
LOT of plates. I use 9g MEA per 500ml of water and that gets me 20 plates. You'll have enough to pour almost an entire case
of plates.
