Inoculating through polyfill?

Would you all recommended inoculating through my polyfil stuffed hole in the top of the jar or opening the lid and inoculating in my SAB? Both would be done in my SAB. Just seeing which one is a better idea.

If you can ensure your polyfil doesn't get wet, then going through the polyfil is probably fine, though not ideal. You should look into using an injection port or getting some high temperature silicone and making your own injection port and inoculating through that.

Make a SHIP, wipe with bleach, flame needle, inoc, cover with tape of a tiny dab of silicone cooties get in even thfough the tiniest hair line cracks in glass so i dont trust an open nip.

I make my SHIPs 1 on top 1 on the bottom for added durability.

Dont stick the whole needle in just enough for you to inject the solution/ culture.

Okay I will make a SHIP on my next jars. The 4 jars I have now with just polyfil should I just open them in my SAB tonight and inoculate them?

That´s how you´re suppose to inoculate with a syringe, open the lid and inject the solution :thumbup:

Poly should be so tight that when you try to inoculate thru it it pushes the polyfil into the jar.

Another issue is that a hot needle melts polyfil.

I don't suggest trying to inoculate thru polyfil tho it can be done

Using poly as a port and GE will work however is more prone to contamination and is a pain in the ass to work with. Buy some RTV and use that as a port or if you want to spend a few bucks buy a rubber SHIP. You can use poly for GE in a 1/4" drilled hole, you may also use tyvek, better yet, a synthetic filter disc.

I've noc'd up many jars when I first started years ago with a single polyfil stuffed hole and had plenty of success (a couple failures as well). However, the success heavily outweighed any failures I had.

Some of my jars still have the same type of lids, except I mostly go from agar to grain or G2G these days. As Bod stated, if you do go this route, you want to ensure the polyfil is stuffed as tightly as possible.

Though, I've never had issues with the needle pushing the poly through the hole or even burning, as I allow the needle to cool for several seconds before inoculation.

Just crack the lid and squirt sideways withiut letting anything (hands or syringe) over the opening. Very safe.

Okay awesome thank y'all for your help. I just knocked up the 4 WBS jars I had with my MS syringes. One GT One PE. How long does it normally take for growth? I'm switching to grain because BRF seems to take a while!

4 to 7 days to see visible growth. 14 - 25 days for full colonization.

I'm getting 3 days to show growth on my liquid cultures on sterilized milo. you'll definitely want to inject with a liquid culture through a silicone injection port

That's just the opposite of what I'd do. I'd inoculate grain with agar through a cracked lid. You should be inoculating your LC with agar anyway. Or inoculate grain with a spore syringe through a cracked lid.

I inoculate my grain jars through the poly with no problem. Less than 1% contams. Just keep the foil on 'til you are ready to inoculate.

I have probably inoculated two dozen jars through polyfill without any issues. Good luck :smile:

Oh okay next I will inject through the polyfil. The 4 WBS jars I did on the 3rd I opened the lid in my SAB and inoculated. No signs of growth yet though!

You really shouldn't be inoculating through Polyfil. People do it, but it's bad practice and it's unnecessary.

For one, how do you clean it before inoculating? You can't just soak it with alcohol. There's a good chance you'll push contams into your jar. Just crack the lid or make an injection port. Silicone is cheap.
