Mycelium extraction to yield psilocybin.hcl

I have read through the posts on here about it and haven't found anything really good, atleast nothing from a true chemist point of view.

I don't know enough advanced chem to know exactly how psilocybin.hcl reacts in most solvents, I can only imagine it is soluable in most polar solvents and insoluble in non-polar solvents like most hcl salts.

Chemistry knowledge required and lab equipment preferable

Here is a basic extraction that should yeild almost 100% pure psilocybin. (not hcl salt)

1 Take jar of mycelium (via a liquid culture, a substrate extraction may be a little harder but would skip to step 3) and strain the water out of the mycelium with cheese cloth.

2 Either vacuum filter the mycelium or use a coffee filter and gravity filtration until very little water is left, vacuum filtration will get rid of 90% of water I would say, so less than 5% of water would be left. (my opinion on that)

3 Soak mycelium in methanol for 48 hours, less would work but be safe, I would say about 100 ml per 10g of dried (12g semidried) mycelium. I would probably use 100g. Shake/stir every couple of hours. Press if you have a French/coffee press.

4 Vacuum filter the methanol/mycelium mixture. Wash mycelium with some more methanol.

5 Boil off the methanol under reduced pressure (atleast 25" hg vacuum) until very little is left, say 100 ml or so. Pour that into a smooth sided jar or glass (easier to scrape that way) and allow the remaining methanol to evaporate. (you can apply a warm water bath if necessary)

6 Scrape all gunk/precipitate from the jar/glass. Should be roughly 2-3g, maybe as much as 5 depending on how much organic trash is in there. Was with small amount of VM&P Naptha.

7 Filter and this should leave almost pure psilocybin, about 500-1000 mg

Complete steps 1-5 above.

6 Dissolve gunk in 50 ml of xylene or toulene in acetone washed heat dried container.

7 Use an HCL bubbler setup to gas the mixture making sure the HCL gas is anhydrous.

8 Filter off precipitate. Wash with acetone.

Should yield about 750-1000 mg psilocybin.hcl

You will need standard lab equipment to do the vacuum filtration and the boiling under reduced pressure. You could evaporate over a warm water bath and do simple gravity filtration but will take longer and be less effective.

If you don't know what a good HCL bubbler setup is, you can lightly soak NaCl with HCL(aq) and drip H2SO4 onto it. You can also make a shit rigged setup like this:

Take 2 quart jars, fill one 1/2 full of wadded up aluminum foil, fill other with damp-rid. Make 2 holes in both lids big enough for rubber tubing to fit in, tightly as possible. Use epoxy or silicon adhesive to glue the tubing in place. Into the aluminum jar connect tubing to filter and run into hole 1. Run another piece of tubing from hole 2 into jar 2 and down under the damp-rid. Run tubing from jar 2 hole 2 into the mixture to gas. Add HCL(ag) via muriatic acid (pool store) to the aluminum, add SLOWLY. Use a clamp to clamp off the addition tube while gassing.

you haven't heard anything, because psilocybin breaks down in acid's like HCL.

It is completely soluble in ethanol, why would you go to a more difficult extraction?

It's not psilocybin-hcl in the mushrooms. It's soluble in hot water, alcohols, and in general polar solvents in its salt form, like you were talking about (i.e. making the solution a bit acidic) or nonpolar solvents in its freebase form (i.e. making the solution a bit basic.)

so you could just steep shrooms in some water, filter shrooms out, evaporate of the water, and come up with some impure psilo* alkaloids?
capsulate the remaining, and your good to go?

people make mushroom tea, and the alkaloids are in the water. i dont see why this wouldnt work, unless oxynization does something.

I think you might get 10mg of psilocybin from a jar of liquid mycelium, if you are lucky.

Yes; I don't see why not, but I don't know how long they'll last. Shroom tea works.

Just not as efficient.
