That is very remote as far as probabilities go. Using a dry verm layer instead of a filter and port is probibly tits up worse than this.

next thing you'll start warning total that .01% of microbial spores will get through his flowhood and could contaminate his projects.



A dry verm barrier is quite effective at holding back contams, I have never had a dry verm barrier fail me.

As for the heating vs wiping the needle down after heating debate.

When you heat the needle everything inside and out will be sterile in fact ever notice when you do flame it red hot it will expel all moisture and turn to a gas thus purging the needle of any left over solution/contams. This is expected since you are quickly heating a hollow tube that might have liquid inside still.

Now take this super hot needle and cool it rapidly, you now run the risk on contams entering the needle where your alcohol wipe will not come into contact with. Thus a super heated needle will remain sterile more often then a needle that now has been exposed to a alcohol pad/time taken to do so.

Sure the chances are slim this will result in a contam but still, its best to inform people with the BEST way to safely inject a jar while minimizing the risk for any contam.

That's why i say a flamed red hot needle will be more sterile then one cooled down with a alcohol wipe, always Flame red hot then use ASAP

Don't know how Shea's logic could be argued. 

I hear of people doing the wipe/flame/wipe thing all the time.  I'm no biology guru, but the following made the most sense to me, Alcohol doesn't sterilize, its a sanitizer.  It only kills via evaporation, and it won't kill everything...every time.  The flame will. 

So why sanitize something that is already sterile?

so i should inoculate with the needle still hot to minimize length of
time exposed to possible contamination.

should i put a bit larger an inoculation to ensure enough spores get
through that havent bin damaged by heat of the recently flamed
needle?

how many cakes should i aim to get out of a ten ml syringe?/how much per jar
i'd be using short/wide half pint glasses with dry vermiculate top and foil lids. BRF & verm. substrate.

To be honest, i have always either did it the way i described...Or i used Agar's square sponge way... (Base Icks posted a link above of Citric's method...I learnt mine from agar about 7-8 years ago, my sponge covers my entire needle and is never exposed to outside air)
It doesnt fail me...
I believe dont fix what isnt broke...
I didnt come here to argue, i came here to spread information...
Dont like/agree with my information, thats fine.  That is your choice. 
This is something most people come here and learn...How to filter through what might/does work for them...

I appreciate everyones input. i like to hear everyones pov. It helps to hear a few ideas pick one that'll work for me.
Think in the interests of science this time i might do one half with a hot needle and half cooled through an anticeptic wipe. (I cant use the sponge method as i dont have any IPA)
Hopefully they'll both be fine and grow without any contamination.

How many cakes from ten cl? Ive got access to plenty of glasses.

(80p for four from wilkinsons!)

Do you mean 10 ml/cc?  A cc = ml.  If that is in fact what you mean, then about 10 cakes.  If you truly do mean 10 centilitres, thats one tenth of a litre at about a hundred cakes.

Hey man noticed your in the uk(Wilkinson's gave it away.) Make sure that the glasses are the right shape, I dont remember seeing "tumbler" type glasses there. You want the short fat glasses, not the tall skinny ones.

Also we can get IPA on ebay. I got a litre for about 6£ shipped. Lasted e about 6 months( I use it a lot), I'm about to order another liter soon.

That's the same thing we said bout wiping before injecting. There are many possibilities like contaminant spores or particles falling into the jar through the layer or being carried in by an insect. The point is that all these problems are so remote as to not being relevant.
