Scavenger Hunt #1 2006This hunt is unlike any of the others I have held so far, simply because there has been such an enormous amount of support from this great community and the very very generous administrators and sponsor/vendor donations.

There are three separate categories this time: one for those who have been here for

6 to 12 months. The category you fit in is based on the day you registered your account.

1) Give a brief description of a spore (it's structure) and how germination occurs.
Answer: A spore is produced from the sex organ (or the mushroom) and is much like a seed, as it contains all the genetic information that will grow and produce the fruit of the mushroom. A spore is a nearly microscopic; sometimes single-celled reproductive body that is extremely resistant to desiccation and heat and is capable of growing into a new organism, produced especially by certain bacteria, fungi, algae, and nonflowering plants.
When spores germinate (reproduce) a thread emerges from the spore casing. When two threads from different spore bodies intersect, they attempt to mate through a hook and clamp connection. A tiny pipe is opened between threads and genetic material is exchanged. The genetically complete threads become hyphae and begin to grow. Spores have four combinations of sexes. Not all intersecting threads are able to mate. Not all matings will produce fertile mycelia.

Answer: Most bulk h/poo substrate failures (other than mold type contams) are caused by substrate BEING TO WET, at spawning. Proper moisture is determined by administering a SQUEEZE TEST (by hand). You want to be able to pick up handfull, that doesn't rip, but does drip a few drops - when you give it a gentle squeeze.

Answer: This myc is being invaded by cobweb mold. When it contacts a mushroom, the mycelium soon envelopes the mushroom with a soft mildewy mycelium and causes a soft rot. It is also a parasite of wild mushrooms. Cobweb mold can be controlled; the control strategies include lowering humidity and /or increasing air circulation as cobweb mold is favored by high and still humidity.

Answer: The sequence of pictures show rye being prepared (one cup of rye per quart jar) and soaked with water for 24 hours to germinate bacterial endospores that sometimes survive the pressure-cooking process.
In the Next step, the grain is strained through a colander to get rid of the dirty water and put into a pot of fresh boiling water (roughly double the amount of the grain). The heat is then reduced to a simmer for 45 minutes up until the first rye kernels begin to open.

How do i make and use an LC from start to finish. (plz be as thorough but as brief as possible, no copy and pasting either).
Here are just a few of the answers received:
-Obtain powdered malt extract, lightest colored available
-Mix with distilled water, one-tablespoon malt extract per cup of water
-Boil water, and then evenly mix the powder until it is completely dissolved
-Pour malt water into a jar with a sealed lid, make a small injection hole in the lid (the size of a typical syringe)
-Cover top with foil, then pressure cook at 15psi for 20 minutes. Allow jar to sit overnight.
-On the next day, inoculate jar with spore syringe through the premade hole.
-Cover the top of the jar with aluminum foil, but not tightly as to allow gas exchange.
-Keep the jar in a warm and dark place, tap the jar a few times every day.
-The solution should start to become milky and near the end of the incubation pieces of mycelium should be visibly floating around.
-Put inside of a refrigerator until needed, the liquid culture is now complete, ready for use
Take a mason jar lid (for the jar you want to use), and punch 2 holes in the lid. Put silicon sealent on both of the holes of the lid, on BOTH sides (This way, your lid on the inside won't rust and get into your LC.) Let the silicon cure overnight.
To make the solution: Get some karo syrup (What I prefer, but you can use honey, and even malt/dextrose, but in this short tek, we won't get into that), and make a solution of the karo and water. The solution should only have 4% karo in it. That is 4 mL karo per 96 mL of water.
Put your solution in the jar, and drop a CLEAN glass marble in there aswell. (For agitation)
Screw the lid on the jar, tighten it aswell, and pressure cook the jar for 20 minutes at 15 psi.
After the pressure cooker is cooled down, take the jar out and swirl it, to make sure the sugars are evenly distrobuted.
Wait for your jar to cool completely, and then using an airport syringe (An empty, sterile syringe, filled with polyfil, and void of a plunger to allow air to pass through), and stick it in one of the injection ports (silicon sealed holes) to equalize the pressure inside. If you didn't do this, your whole syringe would get sucked in. =]
After the pressure has been equalized, inoculate your jar with 1cc of spore solution for every 100 mL of liquid culture solution.
Incubate. When you see thick growth (About a quarter size cloud per 100 mL), you can take a CLEAN and STERILE syringe and aspirate the liquid culture into the syringe. You might have to use your airport syringe to allow pressure equalization.
After you have your liquid culture in your syringe, you can inoculate PF cakes, grain jars, or whatever with it, just as it were a spore syringe.
1) Drill a hole in the lid of a jar (whatever side you may want) with a 1/4" drill bit.
2) Create a 4% solution of Karo corn syrup to water. So that leaves the other 96% water. Put this in the jar.
3) Take a piece of polyfill, twist it up and insert it into the hole in the lid that you drilled.
4) Put a piece of tinfoil over top of the polyfill/top of the jar.
5) Put the jar into a pressure cooker, and pressure cook at 15 PSI for about 20-30 minutes. If you cook it too long, the syrup may carmelize (bad).
6) Leave the jar in the pressure cooker until it cools to room temperature, preferably over night.
7) Once the jar has cooled, remove the jar and clean up the work area, you are going to inoculate it.
8) Once you have a sterile environment, you can flame the spore syringe and whipe it with rubbing alcohol. I prefer to use rubbing alcohol only.
9) Inject 1-2 ccs of spores into the LC, through the polyfill filter.
10) Incubate in whatever incubator you have, preferably at 80-83?F.
11) After about a week, you should see a nice sized globular mass of mycelium. Success!
12) Now all you have to do is sterilize a syringe(via pressure cooking, or buy a presterilized one), and suck up the white stuff in the jar (mycelium)
Now you have a liquid culture which can be used to inoculate anything, for very fast growth, because you don't have to wait for spore germination.
DAY 5
1)What's the life cycle of this pest? How many kinds of them are listed as our enemy? Give 2 ways to prevent infestation...

Answer: The life cycle is Egg-Larva-Pupa-Adult (egg to larva is 4 days, larva to adult 3-5 days and an adult lives for about 7 days); a fly usually goes from egg to adult in only 10to14 days. There are FOUR kinds of mushroom flies listed as enemies (fruit flies and thrips, though listed are not mushroom flies)
Two kinds of prevention are: Farm cleanliness, and a properly filtered airtight mushroom house.

Answer: The best way to text the accuracy of a hygrometer is to test it to a humidity constant, a good way of doing this is using table salt, which when used properly, will maintain an exact 75% humidity in a perfectly sealed environment.
Salt is added to a small open container and a few drops of water are added to moisten the salt until damp. Next, the salt in its small container, along with the hygrometer are placed into an airtight container with care in not getting any moistened salt on the gauge. The starting point should read anywhere between 40% to 80% humidity. Seal the container tightly. Answer: On further mushroom studies you will find that some deadly species from other genera do bruise blue. Just because a mushroom bruises blue does not mean it is safe. Nor that it is hallucinogenic.
