Sterilizing the syringe needle (What is acceptable?)

Would using a stove flame be acceptable, or should I use denatured alcohol as the original PF Tek suggests?

flame the needle with any fire source available until its red hot. Wipe with alcohool soaked cloth/paper/whatever to cool down. Plunge the needle into container, wait 10 seconds to make sure its completely cooled and do your stuff.

Flame it with a lighter till it glows red, then clean it off with alcohol.  In between jars I like to keep the tip of the needle covered in my cotton ball soaked with alcohol.

I usually only flame once, for the first jar.

Seems to work just fine.

Any flame source.. you can make yourself a nifty home made alcohol lamp.. Do to the store buy a like 3 oz baby food jar.. Clean it out aka get the baby food out drill a hole about 3/16 in the lid, fill with 99% alcohol..  Then take a cotton ball and roll and stretch it so that it is long enough to stick out of the jar about 1/2 inch and also touch the bottom of the jar..  Works wonders, no need to hold a lighter...

Also I never swab the needle with alcohol before I inject..  I heat then just inject..  The first rush of fluid from the syringe will cool the needle enough so that it won't kill spoors..

I use denatured alcohol in my home-made alcohol lamp.  The denatured does far better than the isopropyl alcohol.  It comes in a tin in any hardware store.  It is cheaper too.

Flame sterilise between each jar to prevent cross contamination.

You can use any flame source as long as it gets the needle glowing red hot. Regular lighters leave the needle a bit carbonised, but its not a problem. Alcohol lamps burn much cleaner and dont leave carbon deposits on the needle.

I usually just flame it with my homemade alcohol lamp (jar with a piece of cotton going through hole in the lid) and then squirt a little bit out before I inject to cool the needle. I know it may seem like a waste of some spores, but since I make my own syringes, it doesn't seem like much of an issue to me. After injecting into two holes, I wipe the needle down with an alcohol wipe because sometimes there vermiculite on it, and then flame it again, and squirt a little out and inject.

i never flame sterilized or alcohol sterilized my syringes pre-inoculation.  they come sterile and i use a still air box this its not enuf...?

12 fruiting cakes and 24 100% colonized ones would argue :laugh:

The reason to flame between jars is to prevent cross contamination. If you get even a little something on your needle from one jar (or the air), it could infect all the others after. I at least flame between each jar if not alcohol also. You've shown that it's not necessary if conditions are perfect, but it's a quick and easy step to help ensure success.

I just use lighter without wiping. Didn`t have any contamination yet. Will buy alcohol lamp someday though:grin:

nenl, i agree, it is nice and easy,  remember theres no such things as too clean, but i personally dont like the idea of flaming the needle between  jars, because after you start theres liquid in the needle your sterilizing, i know its a minimal amount but i am fairly chinsy and dont like any waste :laugh:

the reason I always flamed after wiping with alcohol was to make sure it was extra sterilized but to also make sure that all the alcohol was burned off. I didn't want the alcohol getting in the jars, but I guess this is not a problem if all you guys have been doing it that way.

ALso, I don't like the bits of verm that sometimes get stuck on the side of the needle to burn on the needle.
