My Elementary Liquid Culture (LC) Tek 9

Damion5050's Elementary Liquid Culture (LC) TekThis is a step by step pictorial of how I prepare my liquid cultures. Please enjoy. Also can be found in my journal. Supplies NeededQuart Jars or Pint Jars Coffee Filter Pot Pressure Cooker Small Bowl Measuring Cup/Spoon Water <-- LoL of course Light Malt Extract Dextrose STEP 1Measure out 2 quarts of tap water and place it in a pot on the stove turn it to high and wait for it to start to boil. STEP 2Measure out 1/4 cup of Dextrose and place in small bowl.  STEP 3Measure out 1 teaspoon of Light Malt Extract and place in small bowl with Dextrose.  STEP 4Once water is starting to boil pour dry ingredients into the pot and stir for 3-5 minutes.  STEP 5Place coffee filter over jar and start pouring liquid into the jar slowly, be very careful and take your time. It is easy to spill it at this point and burn yourself with the boiling liquid. STEP 6Place lid on jar and stick it in the PC for 30 minutes give or take 5 minutes. NotesKeep your Dextrose and Light Malt Extract in air tight containers like in the picture below, if not then moisture will get in and ruin the Dextrose and Light Malt Extract. Once time is up turn off stove and let the PC cool down. I usually let it cool overnight or at least 6 hours before I open the PC. Knock up LC with spores, agar wedges, or a grain kernel from a colonized jar. Also you can either use Pint jars or Quart jars. I personally like to use pint jars as seen in the picture below.

Well the coffee filter is used to strain out the sediment. So u need a filter so if you wanted you could rig up coffee filters in a funnel but this is the way I have always done it.

Looks good D. About the same thing I do, except I went back to using SITR for my Dex and I use a TBS LM to quart. CH

Thanks bro. I use to do karo and honey but they don't compare in a form with dex/lme. sitr is really good as well.
I will post pics here in a week or so when the jars from the tek colonize with mycelium.

Sorry, I dont know much about LCs. I like this elementary tek alot tho.
So if someone were to take a colonized piece of birdseed that appeared to be nice and rhizo, and drop it in there in a sterile environment, would that work pretty well?
Also do you think you can supply the info on where you get dextrose and LME?

Yes you can drop a piece of colonized grain into the LC and let it grow. But like with any LC, after it is ready to use you should always use it on a test jar to make sure it is not contaminated. I always shoot up a BRF cake with an LC to test it out and make sure there is nothing wrong with it.

Very nice tek. I wouldnt mind trying this, but I think I may do it like CH HELL suggested with the sitr. You can get malt in just about any brew store right?
I noticed that this is the second tek you have labeled elementary. Will there be more?

Yes there will be more as I decide to do them. I will have an elementary series. Probably do grain tek then a straw tek then I don't know, maybe a greenhouse tek. Who knows we will see. I started writing this tek like a month ago and finally decided to finish it and post it.
SITR is good, some one did a side by side a while back and LME/DEX was the best in the side by side so I moved from Karo to LME/DEX and never looked back and figured I got this done so why change.

Very nice seems like they are very close. And sitr is free from starbucks so that is probably what i'll use.
Maybe with this recipe I will have better results than I have been experiencing

I stopped using DEXTROSE like you are using and switched the DEX for SITR, it seems to work better but it could just be in my head. SITR is dex/glucose really so it is the same thing SITR/LME lc= dex/lme and like Gangsta said most coffee shops have packets of stir for free. CH

So could someone round up some SITR from starbucks and replace these ingredients at a 4% ratio for the LC?
Also if you dont mind me asking, if someone knocked up this liquid culture with a piece of tissue from a nice fruit, and then mycellium cloud began to grow, how do they make working jars from the master jars?
Do you just suck up like 5cc from the Master LC through a self healing injection port and inoculate into another PC'd LC jar? Then suck up cc's from the working jar to inoculate batches of WBS or whatever it is....and this will be similar to isolating a strain/ more consistent flushes and better fruits.
Thanks alot in advance if you can answer this for me.
