Over a week and still no mycellium?

I Inoc?d 6 ? pint jars 7 days ago, put them in my homemade incubator and still have no mycelium. I keep it at 84 deg and total darkness. I also added two LC karo tek?s about 5 days ago and I don?t see anything there either! Am I just being impatience or should I have seen something yet?
I am using PES Amazonian from sporebank.com, and I?m losing confidence in there product.
If I am impatient, how long until signs of LIFE?

how long after you pressure cooked jars did you add spores? If you didn't wait long enough then it could have been to warm on the inside and killed the spores.

I know different strains can take different amounts of time to colonize, but with the conditions you have,84* and total darkness, I would think you would see signs of life after a week. I just did some PESA and after a week, they were colonizing quite well.  This is a jar of PESA thats been in my incubator for 12 days, same conditions as yours.    Good Luck.Peace,Love,Happiness and Harmony. :heart:Hippie Chick :mushroom2:

It can vary, depending on conditions. Did you shake before inoculating? That will affect it. If its freezing where you are or in between you and them, that could slow things down...or it could just be from some dry spores or a print that just wasnt AS viable. You will see some sort of growth within another 1-2 weeks, and if you dont..then the syringe wasnt viable for some reason. You need to wait at least 14-21 days though..I've had 2 jars take on day 20 before, one from multispore (others did fine, no idea why it took so long but worked fine once germed), one from liquid culture (old culture).

7 days is way too long, the culprit is probably your spore source.

5 hours should be enough but 12-24 is best to be sure...if they were cool to the touch, they should be ok, especially if you shot to the outside...you have to remember though that if the outsides just a little warm, the inside could be much warmer...so maybe you killed a number of spores, some should still survive after 5 hours cool time though..germination might take longer though.

Water content plays a huge part in germination...it can take longer then a week if water content isnt right.

5 hours should be enough, but I would recommend 12 just to be safe.

A week is a tad long, but if your water content was too high, it will/can take substantially longer.

Does the substrate look very moist or slimy?

Well I didn?t really aim for the sides of the jars, just through the holes. As far as the substrate looks? they don?t look wet. They look like the top of a Crumb cake (mmm.. Crumb cake.. ahaghhh..). I did not add a half inch verm layer to the top, just let it be empty air, and I used Tyvek after inoculating.
Because I did not add this layer I can ?slide? the cake up and down in the jar. All the continence of the jar slides together. There are a couple of crumbs on the top, but for the most part it is a solid cake. There is no liquid in the bottom of my jars.
This is the best I can describe the jars consistency. Is this about right?

I don't really know what a crumb cake looks like, heh.

But if it isnt darker in some spots(from moisture) or look slimy at all, I would say your ok as far as water content goes.

Its unlikely that it has to do with the syringe not being shaken enough. At best this would cost you a few jars, and most likely only result in no germination at a few inoc sites.

So, if you don't see anything within a few days, then the only reasons have been mentioned imo.

Only reasons have already been mentioned.

I didn't read all the reply, to many, but if you havn't seen any signs of growth between 5-7 days, something went wrong.

I don't agree with Cray. I agree in part, because it very well may be the case the majority of the time, but I have seen growth start(in some jars) well over a week.

I left a jar in my incubator once that I thought had failed simply because I was lazy and had no need for one extra jar. To my surprise, they showed signs of germination 18 days later. And even still, the full colonization took over 6 weeks.

yea, you must have did something wrong to wait for a cake to colonize for 6 weeks. All depends on the size and the substrate.

Ok, I?m back. I?m going to inoculate 3 new PF?s and 1 LC. I am using Tyvek tape, ? pint jars and only 3 holes per lid.
As far as my old jars go, they don?t look like they are slimy, RootedGrowth. As a matter of fact they all look like the same color throughout. No parts look wet, should they?

If they should, could I inject some sterile water into the dry jars?

After I was done PCing, after 45 minutes, I turned off my cooker, and took of the pressure top. I have a Presto PC and it ?rocks? back and forth. Could the sudden release (steam shoots out for about 60 seconds) of pressure, be the reason there is low moisture?

Or WTF?

I now think they are too dry!

Dude, I was saying quite the contrary.

If they are too wet, it will slow down germination. Too wet occurs more than too dry ime.


Might consider contacting the source as well. They MAY offer a refund or a new syringe.

UPDATE

Well I PC 4 jars last night (one is a Karo Tek) and Inculated them all this morning. I used the PF tek for the 3 other jars + an ? inch vermiculite layer, and Tyvek tape!
I only used three holes per lid, but made sure I was below the Verm layer, and next to the glass before I injected. I sealed them back up with another piece of Tyvek tape!

Is there any good reason these shouldn?t work?
How long until I know?
And does anyone think the old jars will still grow?

I will keep you all updated.
