I wouldn't reuse the plates, but if you are low on them, I feel you and understand why you're thinking about it.
In case it's a "desperate" situation, reuse them, but transfer as soon as possible and keep everything as far away as possible from the other transfers.

Regarding the size of transfer, it can work out even with super tiny pieces of tissues, that's why things like Josex's poke work. But in my experience bigger wedge transfers catch up better, and when doing super tiny aerial myc transfers, sometimes there are no signs of lifeat all.
Do you have pictures of the plate you transfered from?

Hey, I appreciate your input!
No, I don't have any pics, I use pp5 screw on and there is still condensation that would prevent a good quality photo. The thing is that at the same time I made a transfer of a same or slightly smaller size to the cabin which is showing growth, maybe even a day after I saw some tiny hyphae.
But these on water agar have no growth at all, like I've put flashlight from all the angles, and nothing, not even on top of those wedges. Hence why the question.
I was trying to make sure there is some use of them before I toss them to make new ones. And yes I'm low on em. I've only bought 20 of those since that's all I could afford so I gotta juggle with them now cuz I've started a bit too many cultures:muppet:

I took a few water agar clones and they took a solid week before they started growing, but after that they seemed normal (these were pans). I then. Took transfers to 0.5% and 1.5% PDA plates and only 1/2 of them actually took after the transfers.

Give'r a few days to. Recover. Especially if you went from nutrient agar to water agar. Going from food to no food makes me think it might hang tight for a bit. But what do I know, im just a baby 👶

Any of the more science brained among us have book (or anything else) recommendations for getting into the finer details of this hobby?

I know any mycology textbook would do good but if there’s some S tier learning materials I’d rather be as efficient as possible

What would you consider as the finer details?

Micro-biology/bio-chemistry/mycology that is applicable to the mushrooms we grow

Thank you Lo-z, appreciate the help!
I might be able to take some pics tomorrow, I cracked the plates open by 1/4 turn and that helped with condensation. In the process, I dropped one plate and the wedge went flying around the plate:facepalm:

With your  0.5% and 1.5% PDA plates, have you transferred to both at the same time or first 0.5 then 1.5? Was it  1/2 divided between two of those as well?
I hope you're right about your assumption with no food. Btw the cabin one reached the cabin walls today

About a month ago I tested my new liquid cultures on agar. Everything came up clean.

I had my liquid cultures sitting in the fridge on their own shelf without a bag for most of the time since. The needle cap was on the needle the whole time.

Am I ok just to wipe the syringes down with iso and flame the needle tip and then inoculate?

Should I replace the needle?

If you have a new tip I would replace it and flame sterilize anyway. If not and the syringe tip/needle has been covered with the plastic protector, I would first express a little of the LC out to clear any thing in the needle. Then continue to flame the entire needle until glowing before inoculating anything. Just be sure not to melt where the metal meets the plastic on the needle.

For unmodded lids on grain spawn (not pf tek)

After inoculation, do you put foil back on?

And, how tight should the rim be?

I have researched it and found conflicting responses so wanted to see if there was any logic I could follow.

I leave foil on till I get it inside sab then inoc n get rid of foil 😁 for grains anyways

For gar I prefer no pours n substrate I've never tried sterilizing never had to yet

No. Foil isn't needed after inoculation. Just open enough to allow air flow. 1/4 turn maybe.

The only reason I would use foil for unmodified lids is if they were the original metal, 2 piece lid. This is because water pools on them and causes them to rust in the PC. But I don't use those lids. I dropped enough of them to learn my lesson 😂 plastic lids last literally forever, drop them way less, and don't have to use foil.

To be very clear, foil is not a contamination barrier. I also forget that most people here are still using shitty isopropyl rather than elite hydrogen peroxide when prepping their work station.

So just to confirm something because I believe I have been an idiot: Instapots need to be vented too?

Because my dumb ass has been thinking I didn't need to because the IP will do that as part of its cycle. But now I'm realizing that makes no sense because if I leave the valve closed how would it be venting 🤦🏻‍♀️🤦🏻‍♀️🤦🏻‍♀️🤦🏻‍♀️🤦🏻‍♀️

In theory the instant pot vents on it's own by design and should be set to sealing as soon as you start. It should vent all air pockets out before it seals completely.

What are you trying to sterilize?

Yes.  The idea is to purge the air out and fill it with steam, just like a PC. 10 minutes does the trick, then flip the valve closed!

I always play it safe and manually vent mine, it usually gets a good head of steam going 5 - 10 minutes after the pin pops up.

Grain jars. I keep ending up with (usually mildly) bacterial spawn in them and have been thinking through my process.

I vent my instant pot for 10 minutes. What is your process?

I have not been venting. Mine has a "canning" setting that hits 12psi and lasts two hours. I run it twice consecutively for quart jars.

I've also realized that it's having to rebuild pressure for 10+ minutes even if I start the second cycle immediately after the 2:00 clock hits zero. That implies to me it's not even cooking at 12psi for some portion of the 2 hours, so it might not even be long enough 🤷🏻‍♀️

I let it come to normal pressure naturally before taking the jar out
