Made some trenches today, they're not too wide are they? Just making sure. They are sharing a plate because I'm running out and I don't have time to make more right now. I also moved the Iceberg to 2% MEA to see if it wakes up more.

I have to wonder if something is wrong with my agar recipe, nothing seems all that happy on it. I took a new transfer from the dirty germ plate to see if it does any better. I also assume this weak ass growth could be explained by hidden contams so I'm trenching everything that is struggling.

My TTBVI looks strange. It was brilliant white and very puffy immediately upon germination but after some transfers it looks like this. 0,5% MEA

The only one who is happy is my B+ clone.

My knowledge of taking transfers boils down to more whiter=more gooder and I am not pleased with what I am working with  I'm a bit lost, I feel like I'm going around in circles wasting plates making transfers over and over and nothing changes.

What recipe are you using? I had two batches I made with just corn syrup and they all grew like water agar almost. Very light whispy growth but then I used rice water and corn syrup and the difference was night and day. Also when I put the agar from the rice water to rice it immediately started spreading unlike when I put the agar to Corn it took a few days to start spreading on the corn.

i dont disagree with the observation but the conclusion that may be drawn? mushrooms like the same media? is not always demonstrable.

in other words i dont think it matters to the mushrooms.

ime 'water' food sources.. like corn syrup or boil grain waters? are rarely consistent. i like unfiltered brf agar. at around 1.3-2%

I thought grain water agars were pretty reliable?  I agree that the mushrooms definitely don’t really care as long as they have nutrition but some grains are obviously faster then others. But I was guessing that since the mycelium was growing on rice water agar that when I put it to the brown rice it jumped to the grains faster then the corn did and my hypothesis was that since it was growing on that medium already it was used to it and therefore quicker to start colonizing the rice

That is the word around town :muppet: So the hypothesis is that if the mycelium is already growing on GW plates made from the same grains as it will be inoculated into, they should already be producing the digestive enzymes they use to digest the grains.

I have no idea of it is true, but I am willing to give it a shot to see if it matters, I have some species that ain't really fond of grains, but I really want them to "learn" to like it

Right now I'm running different species on 3 different strength GW plates. The only ones moving fast enough to share any pictures is pans and cubensis. Pictures was taken yesterday so this is 6 days of growth.

Highest consentration og GW from left to right.

This will probably only logically be effective for clone culture work. To keep enzyme production already active by keeping that material in whatever it is colonizing. (though entirely unsure abt if agar is considered a different medium than grains evin if there's the same grains in the agar to the mushroom meaning evin with grains in agar it might produce slightly different enzymes?(No clue)) Edit. My 420th post! Gonna go blaze up.

Why would it only be logic to use clone cultures? I was hoping I could use spores to see if I can find a culture more willing to feed off grains :strokebeard:

Is the mycelium even digesting the agar :confused2: I don't have a clue as well, but I'm going to be running this experiment for a while anyway :snub:

Just 2% MEA, 500 ml water and 10g light malt extract and 10g agar. 0,5% MEA for pans. I might make some brf agar again, I just really like how clear MEA plates are. I'll wait and see what comes out on the other side of the trenches.

Just made a batch of agar and gonna throw it in the ip then I plan on using all these to make a bunch of T2s and T3s

Kloakk it's entirely a assumption at this point or oppi ion from me  though why i said clone work would be more logical is because you should be able to notice a difference withen a reasonable amount of time withen a reasonable amount of tests going from spores is a lottery n lwk I have 0 ideas as to if enzyme production and fungal antibodies or whatever they are are a actual trait that is passed on in the spore that could be dominant enough to be effective n doesn't it take a massive amount of time just to select for easily identifyable selections what about enzyme production though... Doesn't seem easy to select for when the second u spawn that same mycelium to a different medium or grain that wasn't what u where selecting for. It will naturally start producing enzymes slowly to balance its ph n whatnot for that medium (I have no research papers or proof of this I clearly state what i think the fungi does and I do not claim to be a expert) xd I might do some proper research tests later for this though i have absolutely no idea where to start for spores to select for enzymes 💀 for clone work it would be a little easier ( I assume) because you can easily make control grows in different substrates to test if the enzyme production kept producing on agar or in lc when transfered to grain or substrate if it colonizes quicker consistently than other relatively friendly substrates or grains to that fungi. I am a noob sorry for the rant ;,(

No problem with said rant and I'm no expert either, but hitting the space bar once in a while would make your post easier to read

At this point I'm not looking for something scientific, just a culture that moves well on wheat grains really. And Darwin is Darwin, if I find a culture that do well on wheat grains it should be possible to take spores from the fruits and hunt for a new clone and rinse and repeat until it's stable on grains, or at least I would think so :muppet:

I’m of this same opinion. I feel like when it goes through it’s life cycle it “remembers” what it grew on and environment etc. so I think it’s possible epigenetics are turned on and that it would get passed down to offspring in an attempt to ensure the next generation’s survival. All speculation tho

Lwk will be a massive project though love it. I'm sure the information will be invaluable for mycologists. I have a similar end term goal of wood chipping cube mycelium. When we figure out how to select for enzymes production or selection forcing to different mediums or substrates il be all in on this research🙏 when u find how leme know
