Hello fellow agar enjoyers, how’s this cabin looking? I tried the cabin on some white rabbit because it looked different then the others and didn’t have good looking uniform growth it had very whispy and slightly cloudy looking growth. I love how white rabbit looks when fruiting so I hope I can get this going. Just dropped some APE into a no pour jar so I’m hyped for that as well. Also did my first transfer of my Jack Frost! Here’s the cabin tho tell me what y’all think

So I tried the agar technique where you place a piece of the donor agar on the petri lid with 2-5 mm between the donor agar on the lid and agar plate suspended above it. The mycelium was able to bridge the gap and the TTBVI mycelium is looking much better!!

Pouring @ around 45c* last night, lucky they didn't clump- but they did set quick enough for this dregs of water agar bottle/ start of ptato dextrose bottle to have made a cool moon plate.

I have agars like this..how many time in months I can store they:

1. out of the fridge at 23/25°C?

2. in the fridge at 6/7°C?

Out of fridge, inside it will get condensation dropping on it, at room temperature probably 6 to 12 months.

I put this vigorous looking plin on agar just for fun and practice while I wait for better stuff to get going. So, considering that it came from a mss T-0 plate should I expect a consistent culture out of this? I cut the stem well above the agar so there shouldn't be any mss mycelium on it. I'm tempted to make a grain jar out of this eventually and see what it produces.

More consistent than multi-spore and very likely to fruit well. But you will still probably have some variability in size, clustering and potency to wean further. Plins are a nice place to start selecting for a good fruiter, but there is plenty more to narrow down if that is your desire.

Thanks. I feel a little attached to these classic cubes I've had from the beginning. Every grow I've ever done is a descendant of my first mss syringe from a random print some dude charged me 12€ for. They can never compete with the superior cubes I just got but I want to preserve them and maybe do something with them some day. I don't know why I'm sentimental over mycelium :lol:

Room temp agar 2+ years revival of chestnuts. I've done reishi lion's mane, shiitake, and cubensis after over a year.

Hello! These are some T2s that are funky, the SNAPE is really tomentose and fluffy. Any advice on these ones to get them good enough to send to grain?




I did a trench on this one, then a cabin, and now it’s been growing out the last two days, but the new one looks super fluffy and tomentose.


This is the transfer after the cabin


Hillbilly Pumpkin look OK? TW x APE Idk? These and a SNAPE are the ones I want to use in my next tubs.

Two Pan Cyan plates after transferring a tiny sliver of just the mycelium that was climbing up the wall of the petri dish. The purpose was to try to separate the mycelium from some sort of red/orange, emedded contam. Credit to Fahtster for the technique. It took a few days to see any visible growth but its looking good now. The MIB is looking the healthiest it has looked so far (over a span of 2 transfers and ~10 plates) so fingers crossed this worked.

My pan also germinated and man it is aggressive and fluffy. I'll have to keep an eye out for the bacteria faht talked about.

Nice.  I’d let em grow out a little more and do it again.  Should get better looking each time.

I will say that since I made that post, I talked to nichrome to get his expertise on what was going on with my plates and he thinks it’s my vabiooth plates that are perforated from multiple pc cycles and it’s actually pseudomonads infecting my agar which tracks with everything I’ve been dealing with.  He also said that several other friends had the same exact problem with those plates and it went away when they switched to disposables. After some research, I read that pseudomonas can discharge different color pigments into the surrounding agar which is likely the different color bands we are seeing.  It was the red in the band that made me think it was Serratia marcescens.

I’ve since gotten disposable dishes and transferred everything to them.  I’ve also started a bunch of new germ plates (those will be what really shows if it’s the vabiooths that were the problem.)

You’re not gonna have to worry about that with the glass plates and I actually had my suspicions about the vabiooth and bought a 5 pack of glass plates a month ago and did some germs on those and they acted like normal so I’m pretty hopeful switching plates cures the situation. 

But that method of taking just the very top hyphae from near the leading edge without touching any agar has gotten me the best results hands down and even if that was the only thing I learned through my whole fiasco, it’ll be worth it. 

My plan now is to simply get clean myc with that method and not worry about isolating sub strains (you can’t be super accurate with that technique most of the time) and then once the culture is clean, then worry about isolating via actual agar transfers

I wonder if HHG cups would have a similar issue?

I'm guessing if one PP5 plate has issues, they all eventually will.

@Faht…would this technique of grabbing the leading edge of the hyphae climbing the lip of a plate result in similar results to mycelium reaching up and growing on the ‘ceiling’ of the agar when the plate is turned upside down, with inoculating wedge on the lid?

I’m sure it’s not something specific to vabiooth but the material.  Here’s the quote from nichrome..

Reusable plastic is made with phenolic binders that eventually denature and cook out of the plastic rendering it water tight but porous enough for bacteria to run through it.



It would explain why all my germ plates suffered the same outcome even from multiple different sources and nothing I did for the last 8 months fixed it, as I was always using the same plates.

I’ll know soon enough but I’d bet the rent on it

Yeah and I believe that’s something ppl do that might have some kind of catchy name like trapeze or smth.  For me, tho, I don’t want to put any dirty agar inside a new plate, even if it’s on the lid.  That’s why I opt for just the hyphae.  Even if the aerial myc does have some bacteria on it, the myc cells are going to far outnumber the bacteria and give it a fighting chance to outrun the bac so you can do another hyphae transfer.. I would think anyway

Let us know how it goes if you try it, tho
