Damn Archespore... good quality pics.... looks great!

I still consider myself a complete noob in all things mushrooms, but got a solid cyanescens agar plate going here! Want to make a master jar to eventually spawn to wood chips outdoors. My timing in the year is pretty off but oh well.

Hey peeps, so a problem I never imagined I'd have...
I've got AGAR GUILT. I'm hesitating to make new plates because I feel terrible about the plates I already have that have died because I did a poor job at cooking everything.

It's silly in a grand scheme thing I know, but it doesn't change how I feel. For the same reason I don't kill anything, I'm stuttering to start again, especially with plates that are unused like b-bar and nec-D because of conflicting temperature requirements. Might have to start up the giveaway again 😘.

Has anybody had this nd' if so how did you shift your perspective to move past it? The whole *sanctity of life as a steward of nature* thing is fkin me up.

AAAUUUMMM - may all eyes that touch this text have rapid and wild success with their endeavours - AAAUUUMMM.

Hey bro, you're a chef making omelettes. You're gonna have to crack some eggs. It's quite alright to just throw plates away, I'm ruthless with my cultures.

Yeah, plates just get tossed. I try to double check if I want to grow it or make another backup (and if so, transfer before tossing). But I often end up with a huge pile to clean up, since use autoclavable dishes that need to be cleaned out, and that's a chore.

Worse for me is when I have old spawn jars I really wanted to grow mushrooms from but can't get motivated to hydrate coir and spawn them. :frown: Or multiple bags of oysters that have fruited inside the bag because I still haven't gotten around reconstructing my gourmet tent. :crying:

How does this sandos pan agar plate look? It's from a spore print .I have done 3  transfer and one cabin transfer. Is it ready for grain.?

Please rate my plates, and I have a few questions I will ask at the end. These were all .5% lme (10 g agar. 2.5 g lme, 500 ml water). These were laid about 8 1/2 days ago.

I am an agar novice and these are the first plates I’ve made. This is going to be kind of long, but I don’t want to shit up the forum by making an entire thread about this.

First 3 were from the same wedge I divided with a scalpel, given to me by another user, told me they were “black cap Natalensis”





Plates 4, 5, and 6 were given to me by the same user, and I were told they are enigma.







Here is one of both plate 5 and 6:



For plates 7, 8, 9, and 10, i used a scalpel to scrape a few tiny spores from a print I received from Panaeoulous co op. The way I did this was very rudimentary and I later learned of the sterile swab method and the different patterns people swab spores in. I literally just dropped a few spores on the agar plate, not ideal here. But I really want to learn to grow pans and the peace river variety speaks to me, as dumb as it sounds, really like the name and I want to work with these spores and develop the genetics. Some of these pictures kinda suck












Now here are my questions:

Does everything look okay?

Should I keep letting the enigma and black caps grow until they cover the entire plate? How long will that take?

The enigma and black caps I’m not super interested in working with right now, however I want to save the genetics via cryovial tek. Now I’ve been reading and realize that t1 plates are ideal. These  would presumably be t1. Would it be more wise to just fruit these, collect spores, produce agar from spores and get a wedge that way? If I saved this, when I laid the next plate it would be t2.


Next question, and most importantly is about the pan peace river.

I realize this is not the most ideal looking agar, should I dump it and start over or try to  do another transfer and go for t1 plates? If not, how long should I continue to grow these plates for? I realize these pictures are grainy, but does anyone see any contam and if So, can I cut around it or how does that work exactly? Part of me thinks it is fine and I should let it go for a few more days, cut the good agar out and try to get some more plates out of it, but looking for an answer from the pros.

Ideally I’d like to have all the pan plates completely covered so i can get as many agar wedges as possible, and that way id be able to make the most amount of plates with the least amount of transfers. Correct?

What you want is uniform growth.



This one is almost good, see how the blue part is different than the red?I'd transfer it and I'd be willing to bet the next plate has uniform growth 360 degrees baby.

The point of agar is to grow out your culture, you're asking how long to do it for, but you should be asking yourself why you're doing it I guess? I'm assuming you want to grow out a uniform culture on agar then fruit it. I'd  Keep a backup plate in case you contaminate or mess something up, for sure.

If I get any contamination on a plate I transfer from it and then throw it away after the new growth takes off. I'm always paranoid it will spread to other plates.

You only need a tiny piece of mycelium to start a new plate, if you're gonna transfer to a new one keep the old one as backup until the new plate is established then yeet it, in my opinion

Once you have a beautiful clean culture then you can slice it up and put it in a jar, cryovials, smash wedges, whatever

The cultures in your first few pics look alright, but the rest are so blurry it's hard to say, but they are scraped from spores so you're gonna want to find the strongest cleanest growth and take a transfer from the leading edge.

With agar, should I be trying to isolate a single strain before moving to grain - like in the RR video - or is it enough just to get some plates without contam?

A strain is 2 single spores so yeah, that's not happening without some serious equipment. That said, only thing you need before sending agar to grains is clean uniform growth :awesomenod:

100% agree on getting clean organized growth and not worrying about isolates, especially as a new grower.  But here's some nerd stuff where I disagree with you Jedi Master Kloakk in regards to isolates.

I don't believe isolating dikaryons is hard.  I think there's misunderstanding of the parasexual cycle though.  Because a dikaryon can have genetics from more than two spores.  And the following example is my understanding of the process.

So, there's three strains in this picture I can isolate with a small enough transfer.  The green is the parasexual cycle.  The red is the resulting cross.  Let's call the strain on the left A + B and the strain on the right C + D.  Nuclei B and D might recombine making a new nuclei we'll call E.  The cross is still a dikaryon, but it has genetics from three seperate spores.  We'll call the cross A + E.  The reason this is called parasexual is because the nuclei do recombine, similar to when a spore is made.  We could even think of E as being a nuclear baby of B and D, as if B and D had made a spore.



Imagine the same thing happens with a heavy swab, but many more times before dominate strains emmerge.

Also, depending on the size of the transfer and the germination method used, isolating dikaryons off a germ plate is entirely possible without any special equipment.  Especially if using germination methods that intentionally seperate the spores and reduce the number of them used like a grab and drag.



Confirming monos though should be done with a microscope.

Isolating dikaryons is not hard or equipment intensive.

Personally I find diverse, cooperative colonies to be more productive and posses more longevity than isolated dikaryons. Diverse colonies have diverse metabolism that is shared collectively.

Select for areas without an internal power struggle. Identify good cooperation, treat it well, and run with it.

I can't find the post of who said it, but I was under the impression that you would need 2 single monos to fuse and that will result in a dikaryon strain.

But how can you be certain you only have 1 dikaryon in the transfer that is being taken? So I thought most of what we fruit comes from several dikaryon strains and this is a variety and not a strain :confused2:
