Good morning!

I am looking for a bit of advice please, I'm finding it very hard to get proper growth on my plates.

They are 2% LME / 2% Agar and I left them for a week before using to make sure they were good.

This is my original plate which is x7x+, from LC i think. Ignore the black bits around the edge, just remnants of a previous charcoal agar which the dishwasher missed and I just re-sterilised ..



This is a larger pin taken from the above plate: there is a some rhizo growth which appears to stall and then what I describe as a "cloud" around it which up to now I've been trying to convince myself is very fine tomentose growth.



This one is a much smaller pin - slight bit of rhizo, plenty of "cloud"



I have also done agar transfers from the same original plate and they all end up looking "cloudy" with no rhizo growth.

My questions are:

- is this "cloud" actually tomentose growth or could it be something else? There are no other bacterial signs ..

- should I be making the agar 0% or just 1% LME to promote rhizo growth?

- should i try a different agar brand?

- is everything ok and i just need to chill? :uhoh:

My first impression is everything is probably fine and you're overthinking it a bit. :smile: Can you get a picture that's well-lit but not from the back?

Less malt extract can encourage more rhizomorphic growth, but it's not a guarantee. And tomentose mycelium is fine too - you're just looking for consistent, uniform growth patterns.

I add a little nutritional yeast (0.2%) to my agar media, which for me makes the cultures grow a little "fuller" and whiter. As always, YMMV, and this is really personal preference - whatever makes it easier to distinguish good and suspect growth, or just makes you happier to look at the plates. :smile:

These two plates are revivals from year plus agar chips. This was two weeks after the dry chips were put to agar and new transfers were taken.

And my first attempt at cabin sequestration.

Finally got back home and nothing on the plates. Either the spores are taking their sweet time given their age or....they no good. :facepalm:

I'll check back this weekend, but if there is still no growth I'm just gonna try a different print. :sad:

Just keeping the thread posted on progress. More waiting....

Grafting tape fixes the Parafilm backing paper waste problem. :lol:

One of the best changes I ever made. Hated peeling the backing paper off Parafilm. (I've tried cling wrap and just hate handling it in general - it only sticks to things I don't actually want it to stick to. Glad it works for other people though.)

Just a drop is enough.

Don't press on the plunger, gently apply some pressure on the syringe body/cylinder. If you use a syringe that is.
