Yeah.  This makes me want to show my mistakes again.

FYI my initial 20 spore plates were pristine until I did my first transfer, requiring me to open the bag the new plates were in.  The outside of the bag was covered in contamination and it covered everything in the box very evenly as all can see.

But I just won't tell the truth next time.  Maybe some other noobs will make this same mistake.

Opening the bag mid sterile procedure is bad sterile technique is it not? Possibly sealing the plates with cling wrap or tape or parafilm will create a barrier blocking the plate from outside contamination?

If you put them in a bag and let the bag get gross, didn’t even try to sanitize it, you’re gonna have issues. You cross contaminated your environment from an outside source and let a contamination in. It has nothing to do with the bag and everything to do with how you did it.

Don’t get mad at trypt for giving you good advice. Look inward and try again ❤️

Just playing the devils advocate here… I’d think using bags with vented plates is probably a bad idea, no?  Kinda like how we don’t use a SAB with soft sides because any movement from contracting or expanding the sealed bag is going to shift all the air inside the bag, even through the vents?

Remember, it was a confession that I made. Trusting the process.  And hoping for support. 

Unfortunately though I was stepped on, in one of my first posts here, by one with thousands of posts.  This is how it is.

I know what happened to my cultures.  And I tried to make that clear for the benefit of others.  But this is just not the right forum for that, and there are some unfriendly people here.  I understand this now and will not expose my weak points anymore.

Don't over analyze.
You asked a question and you found out there was a hole in your sterile technique.

That's great. Take it as progress that you've made. Don't take it as you trying to force conflict when there isn't any. Somebody helped you after you asked a question.

Everything after that is up to your interpretation.

I truly do apologize for being "uncivilized" i cant be "on" all the time. I put a lot of time in throughout my day reading and responding on here, often when i only have a couple minutes.

I no life the shroomery and with how constantly busy i am i often forego small talk and pleasantries.

Maybe its my german blood.

Absolutely. Any time im opening anything i do so slowly without trying to disturb much air. I usually do so from the corner keeping it down in sab and slide the plate out by gently squeezing the edges of the plates.

I wouldnt go that far. A wipe with an iso towel along the zipper and the flaps can remove mold spores and give piece of mind.

Be mad at me all you'd like, i have real problems in my personal world, not to mention my country is allowing its citizens to be deported now..

While i empathize with the lost culture and feel responsible I truly dont give a shit how people feel about me

I would not trust it one bit, I have no problem leaving unwrapped plates in my SAB(with hole covers) for days, but I would never throw them in a ziplock and move that bag around without wrapping them, seems.



I use parafilm and it works wonders, I know some people get old parafilm that starts to crumble and is a PITA, I'm still on my first roll and I must have done +400 plates and probably wrapped them, at least, twice. I keep it in a ziplock and have not had a single issue thus far. I keep my unused plates in a 15qt tub and the inoculated ones in shoeboxes, I probably look and touch them every other day and wipe with ISO before loading it into the SAB, no issue there either.

Call it what youd like, but it never caused a sattelite for me in years. Probably about 4 or 5 now

I know exactly what went wrong with my procedure and I clearly see the forum's 'reaction' to my posts. 

It is evident that several above misunderstand what went wrong with my procedure.  And I will not discuss it any further.  There is simply much anger these days everywhere you look.  I do not intend to participate. 

I will 'have fun with my mold', thank you.

I wish everyone didnt jump in to speak for me and escelate the situation unnecessarily. 

You came in telling people not to use ziplocks because your plate got mold, being an advicate for ziplocking your blanks myself i felt the need to defend the use of them.

I didnt think you were confessing, i thought you were blanketting ziplocks as useless for agar, i felt the need to defend them because i felt bad about your loss.

Im sorry for making you feel stepped on.

Ill give it to ya this might not be the best proof considering i have a ffu finally after a decade, but heres a bunch of vented plates in bags without wrap

No, the only place I keep unwrapped plates is in my SAB, when all condensation has cleared I wrap them in parafilm and then put them in a tub :awesomenod:

And I have no problem believing you do this and do it without failure, but you really know your shit and have been doing it for years :hatsoff:

I'm pretty sure that if I tried to use that technique I would fumble, stumble and fuck it up within an hour :lol:

Thats the thing with practice. You dont learn to ride a unicycle or a snowboard without falling a few times.

Youre telling me you wrap and unwrap every plate before you even put anything on it?

I dont even want to think about how long that would take me.

I have 2 large rolls of parafilm that are useless brcause they are dry and crumbly. Clingwrap is a pita to cut, and

Im far from perfect. I had a grain spawn failure rate last year of like 70+%
Pouring is where my sattelites always came from in my sab
