...and yet another lazier method: generously spray the stipe with ISO and stab it with a syringe full of sterile water, gently squirt to BRF puck (ideally) or agar.

Indeed pokes can work great!  I find them easier to manage before a fruits mature and softer , get them while they are nice and firm

Yeah I think that’s what made it so hard.  I waited nice and long to get some prints.  The ends of the stipes were definitely soft

Alright.  I'm in.  Got my 2 month old spore syringe out of fridge.  Subtropicalis. From reputable vendor so this should be semps.    Knocked up 2 agar plates and just for giggles sent some to honey lc and an old jar of popcorn.  I know spore to grain is not preferred but if everything is sterile I may get lucky.  I just want to get something going.  I can clean up with agar.  I'll post up results here over the next few weeks months

I may not have stayed at a Holiday Inn Express, but I have taken 20 grams of fresh subtropicalis

My plate from the swab I got from lew looks like it might be doing something!

I'm so interested in learning from this species!

Myceliated grain should be grand for gummies right?

the thin stemmed ones are a pita, but doable. you onöy need the tiniest amount. scraping with a sharp blade works for me.
and then there are other lines like „tres soles“, stems as thick as a a sharpie..

As Chris77 mentions, a sharp blade is quite helpful when cloning semps.

they seem to be like Pans in the way that MS performs better, IME, like in the case of this plin:

regretting spreading 2qt by 3 tubs... but I have mushrooms  and very sensitive to light tilting mushrooms indeed

I take the desired fruit and spray it with 70% iso (i know its pretty ruthless) and let it dry for a minute then chop a little piece with a flame sterilized blade (i use my scalpel) and off to the plate (your standard pan agar 0.5%).
Every person has their own way of doing things. Think of me as Granny's way... old and stubborn :heart::oldman:

ps,
This is intended for indoor grown fruits only, outdoors require more attention to detail.

They do indeed, at least in my experience. I just shared how i clone because someone asked. When growing pans, you never go wrong with MS.

Tired of scrolling down to page 4 to find this thread.  So bumping it back up.

Oh.....2 agar plates I nocced up a few days ago  have germinated.  To early to tell if it is the spores or contaminate.  But got some good growth around a visible chunk of spores so hoping at least some of it is semps and I can clean it up with transfers.

What is the current best practice?

I was planning on doing 1 or 2 transfers and then starting a liquid culture for future use and going agar to millet for some immediate action.

I like millet due to the tiny grain size for lots of inoculation points, absolute brain dead easy prep, and anecdotal reports of increased potency.

Still working on my plan once I have clean spawn, but will probably try just coir first and treat it like a cube.......  may do some grain to grain to expand and get some bag grows going.  People seem to have had success with that.

Not in a hurry, just want to do it right and get some fruits so I can see what all the fuss is about.

As with any culture, I would transfer at least once and continue only until you have a clean plate(I've achieved this with other species within one transfer, including a wild cyan print, though I'm chalking it up to luck.)

Then do a wedge to grain. You can expand your grain, just don't run it too far.

I'm unfamiliar with semps as I'm growing for my first time, though I'm unsure how much spawn has to do with potency vs. Genetics. Though I admit I have no studies on hand to prove or disprove this claim, and such a topic would almost be worthy of its own thread.

Though now I'm gonna have to do a jar of rye vs a jar of millet vs a jar of wbs xD

Second flush. I didn’t want to open the tub because of all the fungus gnats. Corn bits in the water are Bti, it seems to be helping.
Time to start over but it was fun.

QQ How long does it usually take after semp fruits open up for them to begin dropping spores? I've got my first successful tub here and some of these fruits have been fully opened with margins curled up for over a week

I just keep misting them 3-4 times a day, waiting

Couple of weeks. You’ll see it once it starts. Post a picture of your tub.

Treating it like a cube your first go isn't a terrible idea, I'd stay around 1:2 ratio personally as well. Don't forget to mist the fruits regularly :rocket:
