Hey guys! How long until you usually get pins in agar plates? I have some 5-6 weeks old plates that are forming “something” lol. The plates need some FAE for pins to appear? If so I might have to remove some of the many cling wrap layers I used

How do you get bacteria in a no-pour setup? I did a BRF agar that's just fine BRF, agar, water. Pressure cook in two layers of tin foil at 17-18 PSI for 30 minutes. Leave them on a shelf still tightly wrapped in two layers of tin foil. Out of 30 plates, one of them had a little bacterial spot when I took the foil off a few weeks later. Since there is no handling, where does the contam come from?

You may not have processed them for long enough, you may not have vented the PC and an air pocket could have formed (rare), the foil had water between the wraps and bacteria slowly seeped in from the outside. 

There's a lot of ways it could have got in there, too many to narrow down from just this.

I had it happen to 2 of mine. I also cracked lids to allow condensation to evaporate, and I'm bad about being gentle with shit. I assumed I knocked them around a bit and something got in. Didnt put much thought into it otherwise. 99% are coming out fine, sitting for plenty of time before use, etc.

It's my first post! Hello everyone!

First one is a lovely A2A transfer of an isolate strain.

Second one is its bacterial brother taken from a fruit, which was my experiment for the past month or so. After four transfers I can't see any bacterial contamination anymore. No slime, no halo, nothing. Still, it grows real slow and has this uneven, bright white and milky look. I want to figure out a solution because I have a different strain in storage that I want to clean.

mine looked like your second picture at first, i gave it time and the thick white blob grew in a nice ropey/rhizo pattern in all directions

the middle calmed down a bit as it spread, but stayed a lil fluffy and remained white

Although this thread is about agar, BRF pucks are awesome for cleaning up bacterial culture. Have a look at the pristine BRF puck tek.

Water is the biggest culprit. Water along the edges wicks up the edges/threads and allows bacteria a way to introduce itself through tight-fitting petri or HG.

Im only 50 plates into agar, and the only two contams I've had were from excess moisture and being on their side (so the water could walk up), which I solved by 2-3 hours in 'the hood'.

Had a brf puck fruit invitro. Put the stipe to grain, and put the opened cap on a plate. I didn't account for the lack of headroom on the plate once it was closed and was unable to flip the cap, so I just shook the piss out of it and am now going to consider all this growth MS. 🤣🤣🤣 There was gill fragments with visible spores on the agar, so I'm assuming a safe assumption. Looks like a mess, but it appears clean so far. Easiest spore swipe so far!

I received a wedge in the mail via this method. Transferred to agar 48 hours ago. Psilocybe Allenii is beginning to take off, in spite of being kept in cold conditions.
