Holy shit

Avoided a major mishap not that long ago -had a media bottle full of bubbling hot agar crack in my hand mid-shake

Still poured those fucking plates though.
(Thankfully I was wearing thick ass neoprene coated gloves and got away scott free)

Damn!  It’s a testament to how tough that glass is.  Mine had cracked almost completely around.  You’re damn right I poured the plates.  Toasted my fingertips through the heat resistant gloves, but that agar got on those plates!

Holy shit

PF Pucks and Cakes
Cakes are all B+
From 1 day after inoculation to 4 days after inoculation:

Albino MVP                      Azurescens                        Tampanensis                    Avery's Albino                    B+

Tub Pins Coming along nicely

B+                                      APE

Might have some pins forming in the bags as well

B+                                      APE

Finally, I don't have cellophane to use at the moment, so I micropored my inoculated plates together.  Nothing to see on these, probably leave them alone a couple of days before checking them.  Will likely cellophane them closed then.

I had a ramble written up, but even I found it hard to read afterwards.  I fed it to ChatGPT to organize for me, and I like the way it laid out my ramble.

Equipment and Supplies
  • Plates:
  • Received a big box of 500 plates.
  • Practiced heating my solid brf agar in the microwave and water bath without breakage.
  • Plan to do a big pour when time permits.

  • Rye Berries:
  • Acquired 10 lbs of triple-washed organic rye berries.
  • Reading different TEKs (Beef's and Bod's) to decide on the best grain prep method.
  • Concerned about the moisture content and workspace required.

  • LC (Liquid Culture):
  • Sprayed some LC onto agar.
  • Have remaining LC in syringes but cautious about contamination.

  • Lids:
  • Using punched holes with micropore for 4oz jars
  • Considering Pasty's unmodified TEK with white plastic lids for Qt jars
  • May use rings and caps for agar
  • Haven't decided whether to try to modify the ring and cap lids

Procedures and Techniques
  • Agar Preparation:
  • Heated solid agar to 190°F in water bath, microwaved to melt, then down to 120°F in water bath
  • Encountered persistent chunkiness (maybe I need a stir bar and heat plate?)
  • It was pourable though, so I'll use this process on my next pour session

  • Storage:
  • Plates currently kept on a warm shelf (75-85°F) due to lack of refrigerator space.
  • Wrapped plates with micropore tape. (I don't have spare cellophane right now)

  • Lighting:
  • Considering adding swan-neck desk lamps for better directed lighting.
  • Exploring lighting solutions to benefit growth without significant expense.

Current State and Observations
  • Colonization:
  • PFpucks and cakes are still colonizing.
  • All pfpucks, including tampanensis, are growing and fluffy.

  • Environmental Conditions:
  • Active plates and grows are kept on a shelf with fluctuating temperatures.
  • No significant issues observed so far.
  • Researching low cost ways to stabilize temps

Goals and Plans
  • Short-Term Goals:
  • Get all cultures onto agar to screen out contaminants and stabilize genetics.
  • Try different agar recipes and perfect pouring and sterile transfer techniques.

  • Long-Term Goals:
  • Reach a point with multiple viable plates for bulk spawning.
  • Find optimal storage solutions and maintain stable temperatures.
  • Explore better lighting options based on growers' experiences.

Challenges and Considerations
  • Financial Constraints:
  • Feeling burned out on buying more equipment and supplies.
  • Looking for cost-effective solutions for storage, lighting, and other needs.

  • Contamination Concerns:
  • Skeptical about the remaining LC due to potential contaminants.
  • Prefer using agar to avoid contamination issues.
  • Will use remaining LC to practice agar inoculation techniques

Miscellaneous
  • Experimental Recipes:
  • Planning to try a clear/colored agar recipe with light dried malt extract.
  • Observations of bacteria-like blobs on BRF plates might be due to chunks or condensation.

  • Future Experiments:
  • Considering in-jar style grows with PF substrate and casing.
  • Potentially breaking up colonized PF cakes into coir for shoebox S2B.

Foamy mycelium can mean:
1. Not enough FAE
2. High nutrient-source tomentose growth
3. Bacterial contam (high heat, high humidity, contam inoculation, contam grain).
This is probably an example of #3.  My grow area sits around 80, and these expensive premade bags were nocced up with older LC from a just-OK source.  They’ll probably fruit eventually, maybe, so I’m just watching and learning while more reliable TEKs proceed.

Culture reserve stack looking good.  Ready to start plating out.

Golden Teacher Contam Rescue Plate

If the white halo is GT myc, I could be in luck!  It's under the agar surface, might be GT myc trying to shed the contam.  Once it's a bit wider, I'll transfer cuts from the edges to fresh agar.
I don't have a GT strain in my collection, it would be incredibly gratifying to preserve.

It doesn't photograph well.  I may need to attempt this transfer sooner than later, since the substrate samples above the agar are starting to look a little green.
It's possible that it's just trich mycelium.  Reviewing the excellent Experiments growing trich thread by Hindsight, it doesn't have that whispy translucent appearance, but only one way to find out!

Harvest and Sampling - 6/9 (nice)

B+ Vogon Tek

150g wet.  Put 50g fresh on a pizza slice, covered in cheese, baked at 350 to melt the cheese.

Pretty sure I inactivated 90% of the psilocybin, because this did almost nothing for me.  Thought the water in the shrooms and the cheese insulation would keep the mushroom temp under 300 - I was wrong

Pulled spore prints:

At the time I thought I just had a bunch of duds, so I ate the caps fresh.  Me Oh My these were not duds.  I wanted to capitalize on the opportunity, having not had any sort of trip in ages, so I ate the most mature, mutated looking APEs from my other grow.

It was a good day

Dehydrated the remainder, which was around 100g fresh, 150F x 24 hours.  Came out cracker crisp and weighing 5g

The B+ substrate soaked in water over night after I roughed it up Beef Style, we'll see if I get another flush.

Next Steps
Agar plates are doing nicely.  I have stack of blank recently poured plates, might do some more transfers today to prep for grain spawn.  Also going to make a fresh batch of agar, might go for a clear malt extract this time.

My PFPucks are doing great, none consolidated yet, will have to decide what to do with them soon.  Tampanensis will need to go in some grain jars for long term storage and sclerotia formation.
Azurescens will need to go in a specialized substrate in a fridge with proper lighting.
The Albino MVP and Avery's Albino and B+ are looking awesome.  Probably do a Tiger Drop onto grain spawn.

Gotta make grain spawn.  I have red wheat, rye berries, and popcorn.  Something about popcorn makes me happy, and I have a feeling the B+ and the two Albinos will gobble it up.  The tampanensis apparently prefers rye, and neither the PFPuck nor the agar plates of tampanensis are ready, so today will be popcorn.

Also going to weigh out the APEs, try to get a spore print, toss the rest in the dehydrator.

Thanks!

Yesterday I did a ton of agar transfers, advanced all the projects.  Didn't take pics of everything, it was a lot, but once there's some growth worth showing I'll do a little roundup.

I've harvested the first flushes of B+ and APE.  Both subs have been abused Beef style  soaked overnight and are set up for another flush.  If I get nothing that's fine, I wasn't even expecting a first flush.  Edit 6/11/24 - already getting pins on the B+

Tried to get spore prints from some APE caps, but as is their wont, they dropped nothing.  I cut em and smashed em and put a drop of water on each one, but I'll be suprised if I get spores from them.  Doesn't really matter, I have other genetics and these were all mutated AF, but worth a try.  I'll toss em in the dehydrator whether they drop any spores or not after the next 24 hrs. Edit 6/11/24 - still no spores, probably not getting any.

Been posting a lot in the Q&A forum.  Really helping me tune my focus.  For some reason I had become enamored with the idea of using popcorn as my first grain spawn, but I've been set straight and will be prepping rye instead.

I have a bunch of 4oz jars set aside, though I'm not entirely sure what I'm going to do with them.  My PFCakes with B+ were inoculated on the surface of the verm, so I just had some sad grey mycelium on top.  I put them under the SAB and shoved all the sad myc down to the BRF layer, will see if it catches.  So I could make a bunch more PFCakes, but I don't really know what I'd do with them afterwards.  Don't really feel like making a shotgun fruiting chamber, I'm pretty happy with spawning to bulk.  I suppose I could spawn them to bulk, or use them to inoculate grain spawn as an alternative to Agar, but I just don't know.  Now that I'm becoming fluent with Agar I just don't know what to do with PF substrate.  Kind of skipped it.

Speaking of, I have three nearly complete PFPucks, trying to decide what to do.  I did the Josex Poke just for practice onto some agar, and also to get some of this B+ strain onto Agar, but now I just have these pucks.  Probably put them into grain spawn once I have it.  Will ask the Q&A if it's worth my time to top them and fruit them.  I'd expect small yields, but since it'll be awhile until I have grain spawn ready and enough colonized agar plates to inoculate them, it might be nice to have a small crop of fruits within the next week or so.

Maybe that's the thing to do?  Have pucks going since they're so low maintenance and just keep top fruiting them to have a low background yield all the time until I'm caught up with grain spawn and monotubs?

Edit 6/11/24 - after talking to Q&A folks, and reviewing the Josex Poke and Pucks post, I can expect these pucks to pin without casing, so I'll just watch em.  The PFCakes still have no sign of recovery.  I'll give em a little while longer, but if there's nothing, I'll combine two doomed projects and dump all this PFSubstrate onto the two doomed sad bags I have and see if that does anything.

My wife spotted a MASSIVE shroom in the compost!

Bad news
She wasn’t able to snap a picture and didn’t harvest it, and a friend who is helping open our pool dumped a ton of floating pool debris into the compost, probably including a squirrel carcass, on top of it.

I hope he picked it for himself at least 🤣

Fingers crossed there’s enough myc down there to give us another shot.  I’d love to catch those genetics, might be trich resistant and have a taste for FLESH.  Any Flesh TEKs out there?  How long would I have to PC a dead squirrel?

Thanks!  I really do appreciate the encouragement.

First a gross picture - I plopped a shroom end onto agar to see what would happen.  This happened.  Haven't researched to see exactly what the fuzzy white mold with black specks is, but methinks it ain't a cube.

I made a streak plate from wiping the mycelium with my inoc loop, and appear to have grown a whole bunch of bacteria.

I had spores on my gloved fingers, so I rubbed them on a plate and got this:

Thinking ahead, I had also used the inoc loop to transfer a clump of spores onto another plate and streaked it, and seem to have grown a nice selection of trich and bacteria.

In other relatively bad news, my second flush of Vogon APE grew a spot of trich, which I tore out.  The blob of bluegreen at the bottom I had thought was blue bruising from when I abused it under the faucet, but now it's pretty clearly trich as well. There are a few hopeful pins in the center there, so I'll allow them to race against time, but won't be lifting the lid indoors.

The Vogon B+ is attempting to squeeze out a couple more shrooms.  I fed it some water in hopes that it would help.  No apparent contam right now, but I'm leaving the lid on until it either contams or produces something harvestable.

My vogon bags continue to consolidate, and the myc is matted on top, so I started spritzing to perhaps invoke some proper surface conditions for fruiting.  Clearly the one on the right is overwatered.  I'll let it stew for the evening and maybe soak up some of that water, then tomorrow I'll drain off what's left.

The first Avery Albino plate is curling up.  The transfers I had taken from it are plenty happy though, and will perhaps be ready for grain when that's available.

The Dirty Teacher continues to look perfectly normal.  Not sure why the bag looked like such utter garbage, but the transfers are doing well, too, so I may be able to spawn and harvest some classic GTs in the near future.

I realized that I had done my josex poke incorrectly - without a syringe of sterile water to spit out the punch biopsy, so two of the poked plates are still blank looking, but at least one looks OK.

Rescued samples of Vogon APE and their transfers are growing slowly but surely.

The FP+ from the lovely Beef is chuggin along just fine, and the baggie has no sign of contam within.  Anyone have a guess for what the little dark circle blob on "B" is?  I may send C to grain after transferring from each for a nice monocolony, only because I'm really excited after finding out what FP+ actually is:

And in "back from the dead" news, now three of my seven PFCakes have come back from the dead:

My cubensis PFPucks are continuing to consolidate, still no sign of pins.

Nothing continues to visibly grow on the "Control" puck.  Azurescens seems reasonably happy on the PFSubstrate:

Tampanensis might be making a sclerotia! Sclerotium?  Scrotum?

I've been hesitant to start making grain spawn for no particular reason.  I can fit seven quart jars in my 23Qt Presto, so I filled up three quarts with triple washed fancypants rye grains and put them in the bathtub for a nice soak.

Goodnight, Rye, I will see you in the morning and almost but not quite boil you for several hours per Beef's Wheat tek

Goodnight to you too, Shroomerinos.

Six out of seven pfcakes have returned from death

Rye is in the pressure cooker.  Hopefully sitting out so long after cooking isn’t a death sentence.  We shall see!

The pfpucks are fully colonized, even the tampanensis is looking done.  Azure needs a bit more time but I’m also not really prepared for the wood and cold tek that it requires.

Excited to move into the next phase!

Considering not putting all these jars to bulk when they colonize, but perhaps instead use them as master jars for g2g.  The possibilities are endless 😊

It was sent through the post office, which means it was squashed flat, shipped in a dreadfully unsterile container, and probably irradiated, and the postal workers opened it up and looked at it under a microsope, called the secret service who taste tested it and decided it was ok to go, then sent it to me.  It was so improbable that it would even arrive that it can't help but thrive!
