I never used colorants before, and I've been growing for awhile (not bragging just clarifying, I realize I'm new here)- maybe I also just wanted to be cool like you guys...

Jokes aside, I tried the black color because I couldn't see the bacteria, and I thought it might show up better with a black background.

I also tried water agar...did some transfers from those yesterday as well. Can post pics if you want to see progress.

I do indeed want to see pics...

Okay, not really sure where to start.

water agar:

transfers from water agar (not looking great):

sandwich tek:

Same cultures, different transfers from t0 (looking okay)

Many of the melmak and PE transfers grow incredibly slow (bacteria), despite what agar formula I am using, some are more or less normal but kindof slow (genetics?).

It's possible I messed up and introduced bacteria somehow into only a few transfers. I definitely have trouble figuring out what's wrong when the contams are not visible.

Try 6g lme and 8g agar for 400ml.

No ny?

I bet my first born he answers: "a pinch"

[gradient:#F5B12A,#]You have little Rustys? awwww <3[/gradient]

edit: I use for 500ml 0.5gr NY for 400ml you can go with 0.4gr
I only noticed with NY that it grows slightly faster. But just using LME-Agar is totally enough. Nutes are not necessary

I don't! That why I can confidently bet my 1st born

I made 9 plates from a swab of APE-R, and got some bacterial growth in 8 of them.

Should I try to save this myc by a transfer, or let the myc outgrow the bacteria?

These look messy, I pulled some cotton off the swab to produce these. They also had bacteria but the myc seems to have taken over:

That growth doesn't look very promising, but if you want to save it definitely transfer away from the bacteria asap. You might get some better looking growth on successive plates.

I'm not an expert tho.

How do you guys feel about this plate? It’s a T5. I’ve been having some trouble getting uniform growth out of it

I would transfer one more time. Personally I'd use 7 and 8 o'clock (and 11 if you're doing 3)

Toss it and start over. You'll have nothing but bad times if you run it as is. 6 transfers is a lot. If I'm not clean and ready for grains by t3 I'll go back to spore

I’m down to go back to spore. That wouldn’t be a problem. Does this look actively dirty to you, or just not symmetrical?

Looks Way to unorganised for 5 transfers, usually its fair to assume theres some of that intermeshed mold that dosnt actually exist apparently lol

It looks dirty to me. The thick clump just below the transfer piece looks gross, the interior growth isn't uniform and that's without even mentioning the deeply scalloped edges

Just start some spores anyway and continue trying to clean, isnt that what water agar is for? To encourage the Desired myc to out run the contam myc?

You know honestly it’s not getting out to grain anytime soon regardless. I’m germing, cleaning, cloning, and storing my library of 40+ varieties. The line of already germed plates waiting to grow is pretty long. I’ll go back to spore.

I think it's been a week, maybe six days. Close enough. Pretty sure I've got some clean myc and as a result, I've been wondering -- when does one start to think about making transfers?

That brings me back to y'all for guidance! I've still not finished the light box so the photos are still meh, and for that I apologize. The condensation on the lids makes things look suspicious but what I can see in person, none of what's going on up on the lids is happening on the surface of the agar, for what that's worth. That condensation is a tricky bastard to contend with... Anyways.

First up is APE. 12 oclock looks interesting, but suspect given its position. Could've been from where the spore solution wandered while wrapping the plate in parafilm. Otherwise, looks like there are some good candidates to go onto T1 plates.

Next is psilocybe cyanescens. This one was trickier to get the admittedly crappy pics of due to the condensation.

My untrained eye doesn't see anything super concerning (aside from the hideous condensation on the lids), but that's very likely due to lack of experience. Any input on where/if either of these plates are viable and or ready for transfers would be awesome!
