PESH swab to MEA agar. The mycelium is growing into the agar, casting a 3d ring around the swab I guess reaching out its feelers in a uniform shape. This is around 100 hours in, temp 26.6 celcius. Is it possible to take a swab from the top of this imbedded swab to agar at this stage or wait a little longer and watch for contam? 30x magnification.

Let it grow out a bit. IME things are still germinating and you could be grabbing all kinds of contams if you grab it now. Let the myc try to run if it needs too.

Sample size is too small to be definitive but here's 4 swab germ plates:

2 on the left are tufts removed by scalpel and burried in the agar, 2 on the right were streaked with zigzag pattern.

5 days in and the tufts have growth, streaks show no sign of growth bacterial or otherwise.



Far left was mangled a bit trying to bury the tuft with my scalpel :lol:

If my recipe is 7gm agar, 7gm ME then how would I make a yeast broth to add 1gm yeast broth to my 400ml media bottle and is it the Yeast that's making the agar trays have stuff in it?

The broth is made by boiling the yeast in the water then filtering before adding to the mix. 

Takes out the solids, but a lot of the nutrition becomes dissolved. 

I recommend a fine grind on the MY if it is chunky to get more disolved.

IMO, take your 400ml or lesser portion, heat the water add your NY and then filter threw a coffee filter, preferably 2x or even 3x. Personally, I would also add the ME to filter that also.

Good job on the cups you poured, :super: but, yes, they are too thick. It won't do any harm, but it's just a waste of agar, time, effort. You should be able to get at least twice that many cups poured. :super: When you go to do transfers you are going to need to shave off the portion you want or deal with columns of agar to place in the new cup.



So the filtering can happen before or after adding the agar and do the ME and NY filtering together before in your example!?

No need to do anything crazy....



My plates are clear as a mofo.



No need for all that filter and pre boil fuss. Add powder, shake, pc...you don't even need to shake it's just an old habbit.

Id do it before adding agar, otherwise it gets too thick to go through a coffee filter properly..I'd just filter out the yeast (if you want to) then add agar and LME.. the LME should come out clear without needing to be filtered

I have only ever had sediment issues with adding nutritional yeast or making agar with flour. All malt extract based plates have been like blue glass.

The clarity of a plate isn’t super important though, is it? You may have trouble seeing the underside of your growth with BRF agar, but for the most part it’s no trouble to have cloudy agar.

Your choice in malt will be the deciding factor, perhaps the agar as well to some degree. When choosing light malt make sure that it's actually light.

Not sure what the terminology is in the US as I'm not a brewing expert but lovibond system for the measurement of beer clarity/colour is one of the metrics for determining the clarity of your beer or by extension your agar or LC broth, you want a 2 lovibond rating which is the lightest.

Your agar doesn't need to be clear, but it doesn't need to be filtered either was my point.

Why add steps?

Water, powder. Easy, but I get that folks like to over complicate even the simplest of tasks.

Precisely why noobs are afraid of agar, and yet the self imposed complication makes it so alluring at the same time.

Yes it's likely the lme.

My broth is just as clear and sediment free:

It’s so funny. I ordered that exact bag from Amazon and they delivered regular malt extract, not light.

But it’s been working great for all my applications.

Next batch of agar I make I’m not going to dissolve it before PCing and see how it comes out. Would love to trim any unnecessary work.
