Re: Dunk your grains before spawning to bulk, post results! [Re: Kenetic]

What do you do if your jars have fruited in vitro? Do they still return to vegetation mode (as opposed to fruiting they are now in) once you spawn them to substrate?

I like to spray some as I mix it up. I don't go crazy tho

Grains lose a lot more moisture during colonizing than you might think.  If youve ever made a GLC, youve probably noticed that you suck less water out than you put in.
Citric suggests rehydrating the grains prior to spawning and I tested this out recently.

I find that:

Colonization times for bulk substrate decrease, sometimes by as much as 50%+.
Fruits are thicker and healthier.
Yields are slightly improved.

There is no added contam risk for a properly pasteurized substrate and fully colonized grain spawn.

Also note that you cannot rehydrate spawn that has not lost a lot of moisture. If you have quickly-colonizing grains (under one week), odds are they will not benefit much from a dunk.

How to do it the quick way:

I take a roll of fiberglass screen (the kind used in window screens) and cut a piece to fit around the top of a five gallon bucket.

Grab the five gallon bucket and clean it out.

Break up your spawn jars and give each a good smell to make sure it is clean.

Dump the contents of each clean jar into the bucket and fill with enough cold water (not distilled) to cover the grains.

Attach the house screen with a rubber band and let it soak for 10 to 20 minutes.

When the time is up, secure the screen with your hand around the edges and tilt until the water pours out. It will drain very quickly, tapering down to a small stream after 10-20 seconds.

Spawn the grains to your substrate as usual.

How to do it the other way:

Break up your grain jars as usual. Open each one and give it a good smell to make sure it is clean. Then fill each jar with cold water (not distilled)
Let it soak for 10 to 20 minutes.

Drain by placing the lid over the top and leaving a gap between the lip of the jar and your lid. Drain it well. The grains will be quite wet at this point.

Spawn the grains to your sub as usual.

A properly pasteurized substrate and clean grain spawn are as contam resistant as it gets, so this dunk can be done with minimal precautions- as long as your grains are fully colonized and substrate properly pasteurized.

If you are using a sterilized substrate, you will need to dunk under aseptic conditions and with sterilized water as well. Do not dunk your grains before G2G.

Some mycelium is lost to the water, but you will see almost no visible myc in the water as long as you dont agitate them while they are soaking.

A commonly asked question is if there is any use for the dunk water after the dunk. While the myc-laden water could be called a grain LC, it needs to be kept clean if it is going to be any use. So if you are using sterile water and working in a still-air box or hood, then in theory you could use the dunk water as a GLC. I would not recommend it though

If you want to make a GLC with the dunk water:

Additional steps need to be taken. Here is a brief outline of the process:

Sterilize a jar of water with a filter + inoc port on the lid.
Work inside a SAB / in front of a flowhood. Use good sterile technique.
Make sure the water is room temp. Use a sterile syringe and needle to move water from one jar to the other.
Flame sterilize the needle between each transfer.
Let the grains soak for 10-20 minutes.
Then shake it up a little and suck the water out of the grain jar and push it into the jar of water.
GLC complete.
Drain the rest of the water off from the grains in open air and they are good to spawn

This is particularly beneficial for species that remain in-jar for long periods of time, such as sclerotia producers. Mycomattie shows the difference between dunked and not dunked tampanensis fruits:

NOT dunked and just crumbled and cased

So I took a bag of grains that had been colonized with an isolate.

I removed one grain before dunking the rest.

I also took a dunked grain of the same size.

I put the two on agar to see just how much the mycelium benefits from being rehydrated before spawning to bulk.

Here is the timeline, today I realized this would be a good thread of its own:

Here is the not-dunked side vs dunked:

Please post results and feel free to discuss and ask questions too :awesomenod:

Tried this and it definitely made a difference!! My only question is would adding gypsum to the soak water make a difference?? or would it be redundant to do so since I do it in the soak before sterilization?

Dunking your grains is not going to do anything except wash off the mycelium, this thread should be deleted.

There's no war, this is retarded information from an ancient thread.

It would help if you guy's didn't venture into threads that even have a warning when opened; learn how to research, think about how or why something would or wouldn't work.

The substrate holds the water, it's the only reason we use a bulk substrate; soaking already hydrated grains is like bringing sand to the beach and then washing that sand with more sand.

In this case you're also washing off healthy mycelium for absolutely no reason whatsoever.

Just think it through for like .2 seconds.

Ancient thread posted by a trusted cultivator. I started dunking my grains and did notice faster colonization but stopped doing it because 1 or 2 days faster wasn't worth the effort

People are not born trusted cultivators, once you post something dumb and then eventually get a tag the dumb post now seems to have been written by a TC.

You did not experience any benefit whatsoever from soaking grains, there is no mechanism to increase performance here.

Correlation does not equal causation.

That's a bit harsh. Deleting the thread would be the exact opposite of beneficial to the community.

Just because the thread is 10 years old doesn't make the info irrelevant or retarded.

Most of my old threads have that warning as well, but there really isn't any outdated info in them, they are just old posts.

Dunking grains is 100% useless.

Cleaning up the org sounds like a great idea to me but I guess the noobs can learn  to think critically instead.

It takes more than one party to argue, so, I'll stop here.

Anyways, you noobs have fun noobn.

Did you do this with the same culture side by side, dunk vs no dunk for control? What was your sample size?

Small sample size but for me it was clear. My un dunked grains wouldn't fuzz up white in under 24 hours and the dunked would. They recovered from the brake up faster for me.

Reasonably isolated, a few transfers, side by side.

Any particular details you feel are important? Like duration of the dunk? Maybe temperature of the water? Anything like that I should keep in mind?

I'm with p9 on this but I'll run a side by side to test this just for kicks

In the end it just wasn't worth the  extra step. I use to run 30 monotubs at a time month after month after month and that extra step added a lot of labor to the whole operation and to shave off one day just wasn't all that significant.

And because I would skip dunking some and not others this caused a lot of over lapping harvest days of which became overwhelming at times.

These days, I've been slacking a lot. Dumping grains into tubs and then letting them sit for too long before I get around to mixing them with coir.

You know what happens?

My tubs end up completely coated with dripping condensation, and the grains start kicking out big fluffy clusters of aerial myc.

Field capacity substrate is more than enough.

Once upon a time, I did the dunking thing. It was pointless and counter-productive.
