Are any of these worth using for T2? They are all fuzzy and not seeing rhizo like I have with B+ plates... Thanks!

Several of those have a good chance of making a more-rhizo T2.

Plates 2, 4, 6, 8, and 10. haha. All even.  And maybe from around noon or 1 of what's showing in most of those.

I'd T from some of those, but also send some of same parts of those even-numbered plates to grain.

6 oclock and 9 oclock of plate 11 looks rhizo-transfering too.

Thank you so much! gonna make transfers soon... hopefully. Works a bitch right now, with a covid patient in facility.

That time of the month again!:

I assume most people here have figured this out already, though I've not seen it mentioned...will post in case it can help anyone...I leave my PCed agar jars in the PC (at room pressure) as I pour. Keeps the agar hot and not hardening.

I tighten the lids (hot!) in the PC, and carry with gloves on, quickly, to pour area in front of hood. Pour, then get another, and put the empty back in the PC, that way the agar doesn't harden and the jars are easier to pour.

This is a PESA culture. Perhaps the PE in PESA now has a new meaning.

Beginning of adventure for first flush!

I swabbed 4 plates with KSSS x APE on 2-12-22.

Pictures below are growth so far.

On 2/22/2022 I did a transfer following @Infinite Monkeys advice taking all mycelium from the largest colony on the blue plate.

Are the remaining plates worth doing anything with?  I have some jars ready to go I could inoculate from a grow kit. (I've smarted up to the grow kit mistake and will be following Bod's TEK from grain to flush)

Does this mean albinism is likely epigenetic and not mutations in the genome?

Albinism is definitely due to a mutation of the genome. Mushroom phenotypes are the product of two haploid DNA sets being expressed in a variety of combinations at varying stages in development and in different physical parts of the mushroom. While albinism is generally recessive it is possible that an albino mushroom could contain one haploid set of DNA that does not have the mutation and thus produce non-albino fruits from a MS grow and possibly even non-albino fruits from a clone culture depending on the nuclear content of the tissue biopsy...or another mutation occurred somewhere in DNA replication causing regular pigmentation in the plate pins. There's an interesting article you might like somewhat related that I've posted before:

Also, a pretty PE MS plate:

What do you see here? I’m on the new side of this and am looking for advice.
Do these look clean to you?
Am I selecting from the right places when I’m taking transfers?
Any insight is much appreciated. Thanks.

You could take transfers from all the plates.

I personally wouldn't advise sending germ plates to grains but if you have spare jars you could send a wedge from one of the plates to a jar or two.

I have a couple of Gt and KSSS plates that Ive begun transferring. Lately all of the myc has looked really thin and weak. I don't know if its my agar recipe, genetics or what but I cant seem to get anything to go Ryzo.

I know its not the end of the world but I'm still not comfortable throwing tomentose plates to grain because of the difficulty of spotting hidden mold riding in the myc.

I'm also getting rings which Im not sure could be caused by the light cycle. They're in a closet with a grow light on a 12/12 timer.

Agar is standard 10g LME, !0g Agar-agar(telephone), 500ML water

These were from homemade prints and swabbed to agar. Should I be concerned? I'm planning on transferring a few more times and cooking up some lower nute agar tomorrow.

Let me know what you all think.

#5 looks super pretty. I would take a couple wedges of that and put them to new plates. They may be ready for grain after that.

Does this look OK for grain?  (light colored spots are light reflections, not bact)

I put it on some antibiotic agar bc I've been dealing with bacterial contam.

It looked like this on some BRF agar with gentamycin:

I xfered to some new (non antibiotic) BRF agar, I think this one was higher nute %:

Wasn't liking that clear growth so I tried it on some PDA and it looks like this so far:

Thoughts? Any of these plates looking ready for grain?

(Side-note): What's up with these warm pours I did? Is this even myc?
