transferring "normal" to "peroxide agar" - dunk the tissue in h2o2 ?

ey!

so i happen to have a couple of "normal" agar plates that are awaiting transfers
to peroxide agar plates and i wonder if i shall dunk the piece of tissue
i want to transfer in 0.3% h2o2 before putting it onto the peroxided agar plate.

what is your opinion on that?

ALSO is the complete agar colonised by the mycelium or just the top
layer of it?
if the whole agar were colonised, like from "top layer till base of dish",
the peroxide dunk would be kind of redundant.

anyway the tissue coming from the plate is gunna be sterile.. you did that one in a flow hood.. the new plate, is all that gunna be in open air? if not you probably dont even need the h2o2 at all...

You can dunk it in H2O2 if you want. It'll only destroy the spores that may have landed during the transfer though. I've done it with clones, plate transfers and transfers to LC with no problem at all.

yes, that is what i thought.
itll be done in open air, thats why the whole h2o2 thing comes into play.

the "master" plate however has been done in a flowhood, yes.

I tend to find dunking in H2O2 to actually work against me, it slows down growth too much and I tend to get more contams out of it.
You're transfering to peroxidated agar anyways, just be fast and clean, you'll be better off the without the dunk, imo.

just transfer the peice to another plate.

When I'm transferring tissue to plates I dip it in 3% h2o2, but I do not bother to do so when transferring agar wedges.

I used to do open air agar work.

just make sure there isn't a draft in the room.

no air movement is key!~

I had very few contams in open air work with agar.

actually i did quite some agar work before,
but always using peroxide agar.

my only concern is that the master plates i am using are "non peroxided"
as they were used to grow spores.

anyway, i will try it with one plate to three peroxided, i hope it will work out.
