Hi Shroomery more noob questions...  Been trying to get a good transfer off a TOC germination plate. I really need to tighten up my spore to agar game.



What do you think about the spots above the areas I highlighted? Thanks in advance and have a good weekend.

Thanks fractal. Isn't getting a clean culture such a great feeling?

I'm on T-6 on a regular cube lol. The yeast and lower lme seemed to help a bunch.

ive been having weird looking myc growth lately and im not sure if its high nutrients or a hidden contam or neither. ive been using the same recipe for years now though so i dont know why it would be growing weird unless it was contaminated or unhealthy or maybe i just took a transfer from a sector that is deciding to grow weird now

is this agar recipe particularly high in nutes? this is for small batches and i just scale up if needed
2.5g agar
2.5g potato flakes
1/2 cup water
1/2 tsp karo

i was thinking of dropping the karo on the next round of plates for lower nutes to try and clean up some strange looking plates

i mean its a run of the mill PDA recipe that i got from pastywhytes tek years ago. its always worked for me. i think he only specified a drop of karo though

i dont have any other ingredients for anything other than PDA agar at the moment as its all ive ever used.

just wondering if there would be enough nutes from just the potato flakes and not add the karo this time?

O Mighty Agar Wizards of the Agar Envy Thread,

I humbly request that thou shareth thy knowledge of the Shroom.

I have but one simple question;

It’s totally cool to poke a hole in the side right?


These are my new pp5s, finally I got some that’s airtight. The black dot is where I want to make the hole for GE(using micropore). Any advice would be very much appreciated. I think it shouldn’t be a problem but I’d just like to double check with the pros.  :biggrin:

You could do but there’s more than enough air in that container for the mycelium to grow. Not needed

The holes are for the PC cycle, I’m pretty sure that it is necessary unless there’s something I’m missing?

EDIT: I’m doing no pours sorry, I should’ve mentioned that.

Nice pics though, is that last one contaminated? What are the tiny little weird thingys?

Oh yeah for no pours the hole will be cool. On the side is a good spot too, no need for foil.

No contam just nutritional yeast. I can't get it to dissolve, maybe I need to try powdered or a different brand.

I'd highly recommend nutritional yeast with LME for your agar recipe, if you're not already. You'll end up using it some point in the future and regret not using it sooner.

I can't get it to dissolve either.

The yeast seems to float to the bottom of the media bottle after a PC cycle/while cooling down.

I have been rotating the bottle to redistribute the yeast while the agar cools.

Rotating the bottle seems to create air bubbles which stay on petri dishes after the agar has been poured.

Next run I'm going to leave the bottle to cool, without rotating it, to let the yeast sink to the bottom. Then I'll pour the agar and leave the yeast at the bottom of the bottle.

In theory, some yeast should still be in the poured dishes.

I don't think it dissolves. You just need to grind it as finely as possible. I weigh it out into a glass bowl and use the back of a spoon, like a mortar and pestle.
