Yea they don’t look too bad to me. Let em grow out a few more days and throw up another pic.

Hey all. So I have an inoculated agar plate. What's the best way to go from agar to spore syringe? or does that defeat the purpose of the agar. I have a glove bag I can use to directly drop the agar into my grain jars but I wasn't sure if that was better. Any suggestions?

Thanks Lenz & D3monic!
Are you trying to make liquid culture? Otherwise agar wedges to grain. Do you have any pics of the agar dish?

Hey and welcome to the shroomery.

I'm not sure what you are looking to do but you have a few options.
First agar is to clean up spores. So what you will want to do is transfer a small piece from your first dish into other dishes until you get nice uniform growth.

Once you get nice clean uniform growth you can either cut a wedge and drop it into a grain jar, then once that colonizes use it to fruit or inoculate other jars. You could also go the route of using liquid culture or inoculate which is either growing the mycellium in a liquid broth or simply dropping the wedge into sterilized water and shaking the shit out of it.

Once you figure out a way you'd like to go with it I can suggest a Tek and give you a good link

That’s how all my edible cultures look rn, lol.

Side note: (@ D3monic perhaps)

Have you ever hole punched a great looking dish but agar isn’t solid and doesn’t quite make the clean circle cut out? It just happened to me and when I tried to pull the circle out to drop in a jar, mycelium is like pulling off from around outside the circle. Idk if this making any sense…

This is what the other plate I’m describing looks like.

Thanks for the response. That's the dish a few days ago. I don't really want to wait weeks/ months for the dishes to grow uniform if I don't have to. I have a very basic home setup. What's the best way to avoid contamination. I have syringes and a spore print but I went the agar route because I read somewhere that it was a safer process.

The spores come contaminated which is the point of germinating them on agar. What you have is a germination plate (aka germ plate). You want more agar petri readily available to make transfers onto.

I’d transfer where I marked in red

Next time maybe transfer a little sooner.

those plates look clean @ BHO

@ Sweetdog that plate looks like a germ plate that's for damn sure
transfer a part on the leading edge maybe at 11 o'clock and there's other spots too if u want kinda hard to see from the pic its poorly lit
best way to avoid contamination - hmmm sterile technique learning to use a SAB and how air moves
also grain prep is huge

You can have a really nice culture that should produce for you from that plate in just a couple transfers if you do it right.

If you fuck it up it's no big deal. Plates are cheap and time is free. Knowledge and experience are priceless. Spores are not hard to get.

These are semperviva/subtropicalis spores that I put on agar 2 days ago and they've already germinated! This is a recipe I am trying out based on one in radical mycology for spore germination. I used 10 grams of cornmeal, a little less than 1 gram of yeast, and about 1/4 a teaspoon of karo to 6 grams of agar powder and 500 ml of water. I also have APE spores that have germinated in the last 2 days using this recipe.

A pin on an albino Louisiana germination plate.

Two pretty melmak plates growing strong. I'm using a hollow metal tube to make these transfers. It's a little bigger than I'd like but it was just laying around.

Nice stuff multifractal! Love to see it, that semperviva and AL especially  :sip: . Been meaning to try out some different recipes for germ plates as well might check that one out.

Thanks Lenz. Yeah, some of these recipes are pretty cool while others are somewhat confusing. For example, I don't understand what place "brine agar" has in cultivating mushrooms but I'll definitely be playing around.
Here is the recipe list if you haven't seen this post:

The complete pdf of the book is easily found online too.

Not to be all mushy (hah), but I actually really appreciate your presence in particular on this forum

What do you guys think of this APE transfer? Specifically note that 12:00-3:00 section where there is some different looking growth moving outward. I see stuff like this all the time in plates and never really thought anything of it but lately am wondering if this is some kind of contaminate?

Man some of those recipes are wild! Like that MYA with cordyceps added, I do not comprehendddd. There's this dude on youtube who runs a farm who's mentioned agar with V8 juice before for stubborn spores so I've been wanting to give that a shot.

I appreciate that dude, and right back at ya. I've been noticing your posts a lot recently and I enjoy seein your stuff so please keep it coming. I'm just glad to be a part of the community and to be doing this cool ass work alongside all the super talented and knowledgeable cultivators on here. Never thought I'd be back to the days of posting on an old school forum like this but nothing beats it

Silkie, anything between like 65-85F is fine honestly, growth is slower at the lower end of that range but I never worried too much about optimal temperature especially for plates. I prefer to keep things on the cooler side for tubs just cause I don't want to make conditions too hospitable for bacteria but for agar it's not really an issue since your cultures should be axenic most of the time.

Hindsight, I would avoid that growth. Not an expert and a little hard to tell from that pic so maybe someone else can chime in here but the way that's growing it looks like a lot like mold whereas the rest of the growth looks like cube myc. I've seen plates where there's a leading edge that's much lighter and wispier but it's usually growing uniformly at the edges of the culture and not like that. That kind of very fine white tangled growth just screams mold to me.

Re: AGAR ENVY! (Anything and All things agar!) [Re: Lenz]

Speaking of the cordyceps addition, I actually made a batch of agar using strained cubensis tea as the liquid base based on the thought that the mushrooms should contain nutrients needed for mycelium growth unless they're all metabolized I suppose. Haven't gotten around to working with those plates yet though.

And I agree, there are some neat folks on this site, especially the hunting forum, whom I'd probably never get the chance to bump into in real life. Pretty quickly one realizes who are and who aren't on that level. It's a little like working with agar. At first it's easy to mistake mold and mycelium but soon enough the truth comes out except in those cases when mycelium and mold turn out to be all jumbled up and unlike agar those are sometimes the best of people while other times, like agar, it's a massive bummer. Sorry for the tenuous at best analogy haha.

Has anyone had issues with water surrounding transfer samples?

Transferred last night and when checked today,i noticed every tissue sample is coated in like a drop/film of water. Most dishes don't have any condensation so it's not like water dripped from above.

I waited about an hour after pouring for plates to cool, then transferred. could that be the issue? Maybe from condensation on agar surface?

Also, i'm curious if you guys wait several seconds after torching blade or just immediately cool into agar plate?

I wait a whole day sometimes longer before I use my plates. I don’t always use all my prepared plates in one shot and have some sitting for a few days at a time.

Also yea you wanna cool into the receiving agar and use immediately. Sometimes I use the top stacked plate that has condensation and use that as my cooling plate.
