Tamp T2 transfers and the T1 donor plate which came from MSS germ plate.
Only 1 plate is looking iffy to me, what do you guys think?

I do 5g agar/4g LME in 250ml of water
I used to use 5g/5g. Haven’t noticed much difference in growth between the two.

any tips with working with swabs?

having trouble with germination.

streaked, removed fragments, dunked the whole swab etc.

still nothing.

I think I have a peu culture that looks pretty decent. Cant decide if im going to drop this one in a jar or not. Anyone have any opinions. I did pull 3 plates off of this one and they look pretty uniform. There is some light condensation on the lids and messes with the image, but for the most part pretty reasonable growth.

Sticking the whole swab into the plate worked for my APE swabs I couldn't get growth from when streaking but if that didn't work for you I think there's a tek floating around here where someone stuck the swab into the agar and then put a couple drops of sterile water on the embedded swab tip.

Might be worth trying, you should be able to find it pretty easily with the search engine.

I haven't worked with swabs enough otherwise to suggest anything else but I'm sure others know more.

Im waiting for PC to arive , but want to start some agar work, I have one litre bottel for agar, sterille petri, can i so agar? Found lots of info, people say it wont work, other it will... Is there eny real good tek?

Working on my first set of clones. I put 3 small pieces of inner stem tissue onto a couple of MEA plates. It looks like each piece is putting out new mycelium, but the growth is kind of weird looking, see the first pic. Can someone confirm this is mycelium and not some mold growing on the decomposing bits of mushroom?

My GT plate looks contamination free to me, but the PESA plate had some condensation on the lid that dripped and spread across the agar and now there are a bunch of bacterial colonies spreading across the surface. When can I try to transfer some of this myc to a new plate?

Agar can be steam sterilized. Id let it go for an hour and a half to two

Ya know, I was super stoned pouring plates the other night (thinking about the honey porridge I was gonna have after) and I reckon I'm gonna see of I can germinate something on stodgy porridge this weekend now.
Wanna try do it without the PC.
Maybe put some in some in small jars in a double boiler type system and then whilst they're steaming put the lids on (kinda like a shitty oven tek where they rely on the hot air rising to keep out contams).
After that let them cool, let a couple go to see if anything grows and nocc up a few.
I'm bored, my cultures are clean or just transferred and all my tubs are in fruiting so why not

Yeah, clone that bitch. Myc will grow on agar no matter how small your transfer is, a young pin grows myc very fast, that'll be strong.

Thanks D3! Appreciate the comment and agar wisdom.

Had a slight hiccup with some recent plates growing mold colonies on the edges, think it might have to do with me being clumsy taking them out of Ziploc's but overall I think agar is my favorite part of cultivation.

Didn't realize how easy it is to generate nearly endless clean cultures once you get a grip on sterile technique, it's pretty awesome. :cool: Now for a flowhood....

Got a couple pins from a Argentinian plate and the growth has been explosive with these bois.



T1 popped off faster than almost all my t7 KSSS clone, although the KSSS have cleaned up very nicely so far! trying to get that perfect even growth all around, so finicky.
