A single swipe of spores will differentiate into dozens, perhaps hundreds of strains on a petri dish. The important thing to remember is that you don't isolate a single strain. You isolate as many of them as you possibly can down to single sectors, then fruit each one separately to determine which one(s) have the characteristics you're looking for.

Remember, once isolated, a strain can be used for many years without degradation by making master slants and storing in a clean lab refrigerator.

for spores next time ill use the "wiping tek" my girl has shown me from her microbiliogy scripts,
prolly use up to two plates for one wipe, that seriously cuts down the number of spores germinating
and makes isolation work a loooooooooooooot easier

btw were way to sober for this board :frown:

It also helps to just barely touch your inoculating loop to the print. However, more spores equals more strains, thus more isolates to choose from. Less is not always better, just less work. It's very easy to go through several sleeves of petri dishes isolating from a single swipe of spores from a print.

how do you know what part of the mycelium growing on the agar to isolate?

the only way to really find out if its any good is through testing?

generally you are looking for the rhizomorphic mycelium,
thats the one that looks like roots.

transfer to other plates and grow out

Yup. The zones to transfer are marked out in the picture above. Use a sharp scalpel and transfer pieces no larger than a grain of rice to get as close to the individual isolate as possible.

Often, it takes several transfers before you begin to see any rhizomorphs. The early transfers are done as much by intuition as anything else.

I have a complete strain isolation tek already filmed for my DVD, so when it comes out you can see the transfers being done and the final result.
