Yellow all,
Thank you for the wealth of info.  Been lurkin and a searchin finally signed up to post. I have an agar q’. Currently the BRF’s (tall 1pt jars... no I did not read the forum well before starting and this is my second go) are colonizing. I ask because my searches turned up that many prefer to let brfs be brfs however I’d like to start some agar ASAP. I don’t see why cutting some myc off a freshly birthed cake to transfer to agar would be a problem. Anyone got advice on why it’s a good, bad thing to try?

Been searching cultivation for BRF to Agar and B2A and haven’t turned anything up recognized as relevant.

Again thank you!  Hopefully I got this right and posted on the correct thread/area.

Why not wait for a pin off your cake, and clone that? Much less chance of contam. . .
My last 5 clones have have no contamination at all, and the clone sample get reabsorbed by the myc. . . More likely to be clean if you tear open in your SAB and extract flesh with a flame sterilized scalpel from the newly exposed surface. . . On to agar, and your golden. . .

So after a week of inoculating my agar with a drop of spore solution I can see a very faint light halo of white fuzz around my drop. It's almost impossible to see with the naked eye, I thought it was still doing nothing, but I happened to notice it when I slightly angled it and held it up next to a window. It's nothing I can take pictures of yet, and my drop is still clearly visible. Is this how the growth starts and will get stronger? Or is this already a sign of something indicative of my culture? It doesn't look anywhere near the starting pictures I've seen where there's an obvious growth of fluffy whiteness and mycelium.

Same.

I've only just started throwing out some that were several months old.

I’m not sure if it will do anything but was going to
Put all my old agar into a blender with a splash of water and pour out into the garden. Same with sub break it up and use in garden beds. Might compost it too. Something to get rid of it all.

That's a good idea. I may try to do something like that. I have kind of a lot now and the idea of just tossing it all out makes me sad.

Look up the Outdoor Growing thread started by bw86 I think-- there are people having success using contammed spawn and old subs mixed in with soils outside--

You make a good point but you can do both.  If your substrate is fully colonized it shouldn't really matter if you're taking a small sample off.  You're no longer in a sterile environment.  Take precautions, sure.  For example flaming off your scalpel and letting cool is probably a good idea.  But really your cake is already fully infected (in a good way). Grabbing a small piece shouldn't alter something that's not already set in its way.

This gets you going with agar which is already beneficial in a lot of ways and once you get pins / fruits you can practice your cloning process.  Win-win!

Yeah,

Thank you for the feedback. Initially I thought it was going to take some time getting supplies together for setting up agar, but as luck would have it a friend of mine was just getting ready to set everything down for a while and lent me everything: an SAB, scalpel, alcohol... had some no-pour agar dishes all ready to go. The agar dishes were prepared some time ago (I suspect a few weeks ago but I didn't ask) only two of them show signs of contamination.

The jars going were all done with a syringe so they have some time to go and are all colonizing at different rates. I held onto the syringes even though I pushed the plungers all the way down. Now I'm thinking of trying to take everything to the SAB, flaming the needle, sucking up some distilled water and making a couple plates. Sure it'll be prone to contamination, but so long as I am able to transfer the clean portions later I should be good to go, no? I figure there should still be some viable spores in the syringes even though the plunger was run down.

Definitely try sucking a few drops of sterile water into a used syringe after flaming (in your SAB)

Eject 1 or 2 drops onto a clean plate and streak. . .

Welcome to the experiment!

made my own thread asking this but I thought maybe this one would be more appropriate. Heres 2 plates they basically sum up my first batch of agar I have some looking like plate one and some looking like plate two. I want to know if either of them show mycelium, and if I could transfer off either of these when they're a little bigger?
Plate 1:

I read your original posting. I'm no expert but I agree with that second reply, the first looks like it might be myc and the second ??? It might help to give a little more context such as the tek used (link maybe) and the source you were taking the swabs from. Either it looks like you got a bunch of particles along in the first frame, or you have something else growing alongside the myc. Not sure what that is.

Very interesting growth on this plate. Looks like mycelium is condensing into a scalpel like shape. Can't wait to get it to grain! :trustme:
