Yes sir myc can grow on top of myc. And i believe there's a few slime moulds that grow white and don't usually spore in agar. They look freaky tho, so you'd likely notice them.

Took another stab at taking some pictures in hopefully better light. Looking for good mycelium growth for transfers. Alot of the plates had condensation that makes it hard to see.

Give it more time. 1,2, and 7 look like they gonna fuzz up real nice. Be able to take nice tissue samples from them. Hard to tell but it looks like you cud right now take a sample from the leading edge of plate 4.

I was wondering what the gouges were, then I realised, they are gouges. . . Pressing a bit too hard with the inoculation loop. . .

Yeah I am pretty heavy handed. Does agar temperature matter? I let them get to room temperature but I think they end up get a bit too soft which is a part of the problem.

Whats your recipe?....sounds like you didn't use enough agar :shrug:

Unfortunately I have more money than time so I bought a pack of pre poured plates of MAE. Don't really know anything beyond that. The transfers I took last weekend seem to be enjoying it though.  :shrug:

I been stickin' with strait MEA for 200 or so plates now and it works great. So does PDA imo.
I'm pretty set with 1g agar/100ml water. But i been dickin' with nute strengths lately. Last batch i used 1g malt/100ml water. I hope it's ok.

I believe the standard recomended amount is like 1.5g agar/2g malt/100ml water. So thats prolly something close to wats in them plates.

I thought most recipes called for 2% agar and nutes so 2g agar + 2g LME + 100ml H2O.

I personally use 1.5% for both agar and nute. . . I copy inski for this. . .

Anyone ever try running many different recipes against each other at the same time?  I prepared the following recipes and did my transfers tonight:
  • LME 1.3%
  • LME 1.3% w/ food coloring
  • LME 1.0%
  • BRF 1.5%
  • Trypticase Soy

Finally took the leap! I got some pre-poured plates to play with initially, as well as supplies to do both pour and no-pour of my own soon. Been following along taking notes :takingnotes: and excited to try this out.

Started a few plates using spores from a syringe (x2) and from a swab (x3, 1 not shown)


Question: is micropore tape ok to seal them? I used that on a few and yellow gasline tape on others. I'm unsure if the gas exchange comes through the sides or through other areas? Thanks

I dnt like using it for wrapping plates. Plus i dnt thnk it does a good job. I prefer saran wrap.

Oh really 2/2/100? Well my 1/1.5/100 was fine so i hope my 1/1/100 will be ok.

I likes the blue food coloring. So does the myc actually.

I thought the blue color made it easier to see growth. How does it help mycelium?

Two of my better looking APE plates, second transfer, brf agar @ 2%. What position looks best to take my next transfer? I am thinking either 10 or 4 o'clock plate 1 and 2. Should I transfer now or let them grow out a bit more?
