Mycelium on agar

A week ago I transfered some spores to agar and they started growing, however I am worried that I got too few spores onto the agar.

My question is if I should try to add some spores to the agar or they are fine like this to transfer on grain when the mycelium has grown out?

Is that where you dropped the spores? If not could be contams. Let it run on, you'll need to do at least 1 transfer anyway so see what it's like in a week then transfer to new plates then if you're happy it's clean you can inoculate your grains.

Should be fine, if that's where you dropped..just wait for the to grow out over the next few days, then take the leading edge and move to a new plate...Ideally you should do 2 or 3 transfers usually imo before going to grain to ensure a clean transfer:thumbup:

Don't add anymore spores. You only need one colony to work with and you have several.

I let them grow to about the size of a quarter or larger then start on transfers. You could go right to grain but a transfer or 2 is the smart way to do it to ensure clean myc.

So far I have understood that with monoculture that it is not certain that it will produce fruits so I wondered that it would be better to grow out as many cultures as possible and then dump themm al into the fruiting medium.

You're not going to get a monoculture by working with what appears to be one colony.

OKay so I understand that I need to trasnfer one colony to another agar plate, let it grow out and then transfer it to grain.

What would happen If I transfered all of the colonies to one jar of grain?
or would it be wise that I transfer all of the colonies to different agar plates and then to different jars of grain and then mix them all toghtere in one monotub?

The purpose of doing transfers is to clean your culture, make an isolate or both. As long as your culture is clean who can use as many or as few plates as you want.

I am really confused right now actually. Initally I though that I needed to just grow out 1 agar plate cut sectors from it and then transfers the sectrors to different grains of jars. Another one of my Ideas was to wait for the mycelium to pin on agar and then transfer the pin to another plate and after full colconisation of that agar plate transfering it to grain.

This is my first time growing mushrooms and since I have bad craving for shrooms at the moment Then I would like my first grow to be fastest and most certain way to go about getting some edible results.

The abundance of different guides about the ways to get my desired results has left me totally confused.
I am really thankful for everyone who has had the time to give me some advice.

OK as long as the mycelium is clean and stays that when then it is almost certain that it produces some fruits?

From MS YES... to further narrow genetics AND guarantee a fruiting culture,  make transfers to obtain a clean culture,  then transfer to grain as you've said, grow out the MS culture and clone a fast finishing fruit from a cluster... you can now take several clones, and grow each out, pick your favorites, and then put those/that culture on a slant for long term storage... slants can be kept in the fridge, taken out and transferred from, expanded and grown... you can work this through A2G or  G2G, and really stretch a culture out with minimal spores

Does not one spore contain one specific genotype and form a colony expressing same exact genes? My worry is that since I have so few colonies on agar then that means that there also are not many different genotypes and maybe none of them expresses right genes to produce fruiting bodies.

Or do the hyphea all express different genes compared to the spore and produce  millions of different genotypes?

All inputs are well appreaciated.

I think you're putting too much thought into it bud. Let that dish grow out a little then transfer from the Leading Edge of one of those colonies away from any contaminants if there are any. Grow a new dish out to about the size of a quarter or half dollar then cut out as many wedges as you like from leading edges and drop in clean Grain jars.

Too much thought, 2 spores make one dikaryotic culture... but unless you're in a lab and just experimenting, it'd be impossible,and there's absolutely no need for all that... grow out, pick some clones, grow them out, pick your favorite and slant it

Gonna be starting mine next week, for sure gonna clone my current set once they start pinning... might even just take a pin and throw in on the agar, then transfer a few times.

That works... a little quicker to monoculture, but you'd be blind to end product until the second tub grew out... :shrug:

yeah, don't get me wrong, I will also to tradition cuts and clone as well as some MS. seeing as pins on agar are gonna contam in there.... Just wanna try a lil of every way to culture since I'll be just starting out.
edit: ahh I see, so even though I'd agar a pin, it wouldn't reflect other fruits in the tub.. gotcha, gonna just cut off the biggest boy in there once they start then tehe

Id wait for a fast finishing cluster fruit... take clone from inner stipe tissue to ensure cleanliness... you'll have to further isolate the culture either through transfer or additional clones, as many single fruit will still contain multiple strains performing as one... I prefer the monoculture  (multiple strains acting as one) than the isolates I've separated...
