Rye Contaminated EVERY TIME

Every time make a batch of rye it gets contaminated. I use organic rye berries and distilled water. The ratio of water to rye is good and so is the total volume in the jar. I use quart jars. I put the rye in, then the water, and rubberband a piece of aluminum foil over the lid. I let them sit for 24 hours and then PC for one hour at 15 psi. After they cool, I quickly and cleanly remove the foil, shoot some solution in, and put it back. Then they go in the incubator at 83 degrees F. After a few days, they get some cottony patches. Then, the jars start to get that sickly sweet smell that can only mean bacteria. The mycelia stops growing and I get very sad. What am I doing wrong? Is it my foil technique? It seems kinda prone to contams that way. Should I be using filter discs? Where can I get them?

Yeah dude, I ve never heard of tinfoil being used as a filter disc, are there holes in the tinfoil or something. I use two coffee filters on the top of mine with the lid on top of that. I punch an inch in diameter hole in the top. Are you sure your water ratio is good. I use 200ml water for every cup of rye. I forgot to mention I am using normal rye because I can't find rye berries. One more point to add are you sure tou let the rye soak for at least 24 hrs, That makes all the bacterial endospores come out of the rye, any less and you could be inviting contams right into your own home.

1. Prolong the sterilisation time to 80-90 minutes.
2. The soaking time(especially now in the summer) might be too long. Reduce to 12-15 h(over night).
3.>Should I be using filter discs? Where can I get them?

Get some filter disks as in the post above. Prep a few 2 hole plastic lids. Soak rye overnight, heat it up a little before you plan to use it. Don't boil, decant, rinse well, drain well in a collender.

Load jar pints or quart jars 3/4 full. If they seem to still have to much moisture, nuke them for a few minutes to steam out some of the moisture. Apply the filter disk, lids, rubber band triple coffee filters over the lids & tight to the jar. Leave the lids a half turn loose.

PC @ 15 pounds for 70 -- 80 minutes. When the PC cools a bit, remove & shake jars & tighten lid without removing the coffee filters. Store in a clean place until cooled to room temp. With all sterile precautions you can muster, remove coffee filters & innoc through hole in lid -- right through the internal filter with what is hopefully a clean -- viable syringe, then swab clean, tape over the innoc hole & replace coffee filters.

Incubate jars around 80 -- 85 F in a dark clean place & you should have some better results appear in 3--4 days. Don't inspect them to death. It doesnt help the myc colonize faster .

Are those the kind Stamets sells at Fungi Perfecti?

My cat's more fond of the ones you can get at Micron Magick. They're a lot cheaper ($15 for 100!) and kitty's never had a problem with them.

The kind Stamets sells are great, but too spendy.

Micron Magic disks work as GREAT. The Racoon used 200 of them & never had a problem.

Then he stumbled into a source of similar, but thicker spun filter material in rolls that are 4 feet wide & 100 feet long for about $20 bucks. Now he uses that instead, because it is easy , he gets the rolls localy & even less expensive than either of the above.

The Racoons understanding is that the optimal temp for spore germination & myc growth thereafter is 86 F. The Coon has had great germination & fast colonization with temps ranging from 72 to 85F w/rye, wheat, other grains, milo, red, white millet & bird seed mixes.

The incubation chamber (uninsulated 5 drawer tall vertical suspension metal filing cabinet) has about a 5 degree temp fluctuation from day to night. So, the average ambiant temp inside is close to 80F.

By the way, RubberMaid 4.9 gallon containers fit the shelves of that filing cabinet 3 wide to a shelf (w/room to spare between them) and the whole set up is inconspicious & neat. Works great for colonizing those small tubs of substrate on a few shelves & jars on the others.

If the Coon would get off his lazy ass, he might post a pic later.

I feel your frustration. I use the same prep methods in "Let's Grow Mushrooms." Trichoderma is very frustrating. This is how I minimize it.

I use rye berries and WBS. I inject spore, LC or transfer agar in a closed air chamber, alcohol prepped and with UVC germicidal lamps to kill bacteria. I UVC my bags, agar, spore syringes and prints. Are you using ball jar lids with a breather patch and injection port? I would not merely place foil around the top. I also do not know your methods of sterility.

Peek at the rye bag pic attached. Colonization after only about 5-6 days using Cambodian. When I use agar or LC my mycelial growth is twice as fast.    All conditions are controlled.  I rarely get contamination. Almost never actually. Is your source for sterile spore syringes reliable? Could they be contaminated?

I incubate my spawn at 78° F. No higher. PM me if I can help you. I think we can over come this. I also have prints, spore syringes, agar,LC and colonized rye guaranteed contaminant free. Let's get this contamination problem under control and see about sending you a good spore syringe or some strong LC. Keep your chin up.

I know what your saying and respect that but I had an option. Buy a spendy laminar flow bench or for under $120.00 I built a very effective UVC chamber and my contam rate is practically nil. I saved a lot of money in the process and my boomers are awesome. Serious results done very cheap.

I just use a still air box, the uv thing is overkill, flow hood are overkill too unless you're making 100s of jars a week. or just want to spend the money

It may be overkill using the two 8 watt UVC's but the cost benefit analysis, so to speak, resulted in me buying two $20. lamps from Walmart and 2 UVC 8 watt bulbs online for under $15.00. My box appears so sterile that after inoculating 30 petri dishes of agar with LC, I once again have a zero contam rate. I feel the UVC has paid for itself repeatedly.
