Pouring & Using Pre-Made Pre-Sterilized Agar??? (HELP)
    #6661318 -

I've been all over the agar archives & they left me a bit confused on some points.

First of all I got the "Basic agar culturing kit" from Spore works and this comes with a jar of "solidified" Potato Dextrose Agar, I guess "jellied" is more to the point. In some of my reading it was said that nutrients needed to be added to the agar, and I needed to PC it.

This is a PDA that came to me pre-sterilized, and I was under the impression That all I had to do was heat the jar in boiling water until it's molten, then using sterile techniques (homemade glove box in my case) pour into the supplied petri dishes then I was good to go

Needless to say there were no instructions with the kit ,nor did I expect any, I figured I knew the basics and what I didn't know I'd get from this site until I hit the Archives. Please, any clarification would be greatly appreciated.

Thanks in advance

-P.M.-

Lets see if I can shed some light on this for you.
First off, what you purchased is ready to pour once it is liquified. Just make sure to adhear to a sterile technique.
PDA has Potato dextrose in the agar already. Just like if you had Malt Extract Agar. Agar Agar is just agar.
The kit you bought is ready to pour, you all set to keep learning.
IMO if you have access to a PC I would recomend Mixing , Sterilizing, and Pouring your own. Much more cost effective. Just my opinion. Ask if you would like more info. I might do a whole agar tek on here for you and others.

Thank you! I've been sweating bullets waiting to see if I would get any answers at all.
You pretty much told me what I wanted to hear. That's what I thought since before buying the kit, but sometimes the Archives can be a bit contradictory.

And an Agar Tek would be terrific, I'm sure there's one already out there somewhere, but not here, not now.

I saved all my old potatoes and couldn't find a Tek to use them with. So their gone now.

Thanks again,

-P.M.-

Piece of advice, to remove all doubt, buy or borrow any of paul stamets books, preferably The Mushroom Cultivator, or ( I think this is the title), Growing Medicinal and Gourmet Mushrooms. The first one I mentioned you can count on as THE bible of growing fleshy fungi.

Once you hear so much about any one thing, be it a movie, a restaurant, or, especially a book, it's time to further investigate said "thing".

I've heard enough.

I'll have the book ordered by tomorrow!

Thanks,

So I poured the agar in my sterile glove-box, I had just a small amount of agar (enough for 1 more petri) left over. This was 2-3 days ago and no signs of contam. So I guess all's well.

Thanks for the help,

Now the question is where to go from here.
I have so much going on that I'm not really able to do too much research (at the moment) on how to inoculate the agar with the fresh shrooms that I am currently growing.
I know that this is a better route than multi-spore generation, but the actual procedure is still a mystery to me.

Of course in time I will do the research and figure it out on my own, but any help would be much appreciated.

Thanks,
-P.M.-

To prevent eventual contamination of your plates you should wrap the edges. You can buy Parafilm but its expensive. I personally use Glad Cling wrap mini rolls and stretch them over the exposed edges to seal. Mini rolls are 2 inch rolls cut from a full sized roll of wrap. I do this immediately after cooling and redo it after inoculation with spores or tissue.

If you want to attempt a clone of a current grow, choose a nice young healthy mushroom and cut off the dirty base. I douse my hands in alcohol then split the mushrooms from the base to the cap with my fingers. Using a flamed and cooled scalpel, take out a tiny bit of clean tissue in the very center of the exposed split stem. Place on agar and seal with wrap, incubate. You should see it fuzz up and move onto the agar within a week or less. Any slimy material is bacteria or yeast (bad) but if you have white mycelium growth as well you can attempt to move that to a clean plate. Good luck!

I sealed my freshly poured petri's with electrical tape till I ran out then I went to masking tape.
I figured that as long as it was sealed; then it was sealed.
I recently found a roll of breathable medical tape, which I originally intended to use, but I see now that it would serve a better purpose after adding the tissue to use as a sealant.

Now when you say incubate, you do mean to put into my incubation chamber as if it were a jar, correct?

I sure appreciate the quick responses from you guys.

Thanks,
-P.M.-
