i started agar this weekend with my new flowhood. poured me my first plates
where do you put everything? what stuff do you have in front of you when you're cutting and pasting? where do you put your tools, your alcohol lamp? do you have a shotglass of water or alcohol or peroxide in there? and whatcha got? what's an inoculating needle? and what is the bent wire for? do you rest them on anything?
tips me up

When I am pouring agar, I have a stack of blank dishes infront of me, and a stack of filled ones to the left of that. I take the top off of the blank plate, place it to the right (bottom-down so shit can't fall in it), pour, then place the top on and put the filled dish on the stack. You want to keep them in 1 stack to minimize condensation.
When I am cutting wedges, I don't use an alcohol lamp, I use a MAPP gas blow torch, some curved forcepts and the blade from a scalple. I pinch the blade in the curved forcepts, so the when the forcepts are set down the blade is held in the air (by the curved section) and never touches the ground. I only sterilize once between a few plates, assuming I am working with the same isolate.
To cut and transfer, just use the blade for everything... cut a wedge, scoop it up and stab it, then place it in the jar and tap the wedge off the blade. You don't want it to touch anything except grains.
Innoculating loop is a little loop made out of thin wire, for innoculating spores. You sterilize it, swipe it on the spore print then touch it to the agar (you wont see any spores.) I don't use a loop, I use the tip of a syringe because it's easier for me, I don't like how the wire bends so easily.

I blow torch them until they are red hot, then put them under my hood. I wait for them to cool, it takes less than a minute. I put the plates in a plastic baggy, they're always sterile due to the processes used ot make them. I store them in a plastic bin. You put something on top of it which is hot, while it is cooling down, and don't let it cool down too fast. For storage of tools, I keep everything in $1 "shoebox" plastic bins.
