What do I google to get one of those fancy bottles yah got there?

I was just using an old full size 1/2 pint jar I got from when I bought premade jars.

for grain soak agar probably yes
for most other agar recipes with amino acids and some sugars you don't want to cause maillard reactions and you also don't want to caramelize any sugars.






If it was going to caramelize, it would do so within those 15 minutes, so I would say that a longer PC time is just fine.

Considering even the toughest endospores get killed in 15m in liquid media I would say you're fine but do what works.

They have a special removable ring on the top of the threads. The pour ring is specifically to not make agar dribble Down the jar, it pours out like it should

Yup! I love the lip ring feature! Makes the pouring a breeze with no drips at all! You don't have to give the bottle a twist at the end of the pour to prevent that last drip that you would get with another vessel. When I first read about the lip ring, I didn't think it would make such a difference. But after using it, wow! These bottles are totally worth it.

They are really easy to pour. I find the glass above the agar level feels cooler to the touch when pouring than the cuervo bottle I had been using. The cuervo bottle would drip after every pour unless I gave it a shake at the end of the pour to get that last drop onto the plate. I highly recommend the media bottles. Hell, I'm excited to get home tonight to pour some more plates with my new media bottles! I just received 11 cultures and I got some serious work to do. LoL!

Hi Bodhisatta,

Thanks for sharing those lovely little Pyrex Media Bottles.  I'd like to buy some.  I was wondering if you could answer one question?

1.  Did you drill holes in your lids and insert a synthetic filter inside like RR does in his video and then cover up your lid with foil before putting in the PC?

So I do not need a hole in the bottle cap of the pyrex borosilicate media storage bottle. Thank you.

This will be my first time doing agar.  I'll be placing my bottles in a newly bought AA921

I'm curious how much (agar mix + volume of water) do you put in your 500ml bottles?  I was under the assumption that I needed to have some headroom in the bottle - stated differently - the bottle should not be filled to the maximum capacity of 500 ml.


I was trying to get more information because I see that you, Pastywhite, Juiceh, and Stromrider are all using the same bottles, while most other people have chosen the liquor bottle alternative. 

People who use liquor bottles usually place a synthetic filter or polyfill inside the lid to trap contaminants because of the vacuum created when the agar cools once the bottle is taken out of the PC.

I fill my bottle to 400. Which does 20 plates once you're good at pouring.

Some old hands do the filter lid. I used to. But I got a job in a lab and saw how everyone else but these mushroom guys did it. Since then I just used normal lids and my success rate has gone up. Not because of the lids but because of practice. The lid made no difference so why modify it.

I'll follow your lead and fill the bottle to the 400ml mark and then practice pouring in the SAB until I get 20 plates.

You can practice with a sleeve of dishes but you waste a sleeve since they won't be sterile anymore. It's maybe worth the practice if sourcing dishes isn't hard or expensive

A sleeve of 10 polystyrene dishes is $4.00 where I live.  I've opted for a case of 500 (90mm x 15mm) dishes for approximately $70.  I think the practice will definitely be worth it.  Thanks for the suggestions.

Quick question - what magnification range do you suggest on a microscope? Id like to buy one for use with agar to help insure clean samples and for fun.  Ive been told Dino-lites work well.

400x to see spores barely bacteria. Phase contrast at 400x helps for bacteria without stains. 1000x oil for seeing bacteria and detail in hyphae like my pics of clamp connections.
